I am preparing quart sized mason jars for rye colonization and had a question about the inoculation. When preparing the lids of the jaws does one need to use tyvec or polyfill necessarily? couldn't you just poke two holes in the lid like BRF? And doesnt the big hole stuffed with polyfill allow a greater chance of contams because it is a bigger hole? Also when using the polyfill method how do you inoculate?? Thanks a lot!

You need a filter, not open holes. Some people shoot through/past the polyfil, others add an injection hole or injection port. I used tyvek for a while and went back to polyfil. It makes a nicer lid, in my opinion.

so when inoculating just stick the needle through or past the polyfil? The hole that is about the size of a penny that is stuffed with polyfil does not let in contams?? thanks

Yes. Or you can use a second, smaller hole. Just big enough for the needle. Take off foil, shoot, put tape over hole.

Awesome, thank you =]. No need to put another lid or tape over the polyfill when sterilizing right? After i shoot the jars leave em as they are and put into normal incubation temps? Or should i cover the polyfill hole with tape? Also would there be another way to sterilize the room i am working in when inoculating besides using lysol because that stuff can't be good to breath in. I was thinking a hydrogen peroxide spray or something? Not sure of ratios though ha. Unfortunelty i do not have a Hepa at the moment so i am going to have to figure out something. Got any ideas? Muchos gracias

Foil over the top to keep it dry and clean until you shoot. Then nothing after you shoot- take the foil off to allow gas exchange. Please forget about peroxide, it has almost no use in this hobby. Likewise bleach- I don't use it at all.
IF you are uncomfortable shooting in open air, make a glovebox. Don't fuck around wasting time with sprays or other crap.
