Liquid culture jars..

hey i was thinking about buying an LC jar from mycosupply, do any of u know how long it takes for the spores i inject into it to germinate? idk, i always just break a piece of a fully colonized spawn into a jar and shake it and just use that, i just wanna make sure my jars completely sterile so im just gonna spend the 11 bucks.

This is just from personal experience so take it for what it's worth...I have never seen a LC from spore take more than about 3-4 days to germinate at room temp. An important thing to note though is that you do need to have the jar depressurized (either by opening in front of a hood or by a blank syringe filled with cotton/polyfill and soaks in rubbing alcohol). Those are my two cents.

3-7 days, five on average, using MS.

no i do have a PC but to get the piece of spawn in the jar to shake it i have to take the jar lid off and put it back on quick before too many endospores get in. i cant remember what the tek was called. i just want a way to be absolutley positive my jar is sterile and i dont think thats realistic if a tek requires u to take the jar lid off after sterilization.

Since you have your own PC, make your own LC clicky. It's possible remove the lid from a LC in a glove box or even easier and safer in front of a flow hood. If you don't think it's realistic to take the jar lids off after sterilization, how did you plan on aspirating your LC into syringes? HL

i dont have a flow hood, ill check into them. and i never felt like making a glove box, i know should anyway though. and i just flamed the needle till it was red, swabbed with alcohol and i only put 1 hole in the jar lid and stuffed it with poyfil so i would just stick it in there and suck it out like that.

Polyfil for a GE filter on LC's is a bad idea. When using polyfil for the GE filter for LC the risk for the polyfil filter to become wet and wick in contaminates is a guarantee. You have to shake LC's at full colonization to break up the myc to even be able to aspirate the LC into syringes in the first place. How are you gonna shake your LC up and not get your polyfil GE filter wet? Polyfil for GE is best suited to grain jars. HL

i mean i knew it would get wet, i guess i thought since polyfil is inorganic the contamination rate would be increased alot, but it was a while since i last did an LC.oops

If you wanted to depressurize the jar take a syringe, remove the plunger, stuff it with alcohol soaked polyfil or cotton swabs, then inject the syringe through the self healing injection port and and the air get sucked into the jar. Do you have any filter disks?
I remember finding a link on here for really cheap hepa filters and I just made the box for the hood myself and used a squirrel cage fan as the blower. I'll see if I can find it again.
The way I do a grow in general though is to use a lid with either a filter disk or tyvek over a whole in the lid. The lid also has two-three holes filled with silicone to act as self healing injector ports. I then take a sponge and soak it with alcohol, put it on top of the injection site and take the spore syringe and inject into the jar the amount of solution you want (.25-2 cc [my preference]). This eliminates the direct need for a flow hood. You can then spawn to a bulk substrate or do grain to grain transferes. You'd need a flow hood or glove box for the g2g though. Don't know how much all of that helps but lemme know how it goes! This is a really fun hobby once you find your own preferences and techniques.
