infected syringes

Hello all,

I could use a pointer to the hot agar over an infected plate issue.

I just need to confirm the temperature (and how to meet it) that
the pour-over needs to happen.

Backstory: I tried making LC of four new varieties
(Elephant Dung, Cambodian, African Transeki, and Jalisco).
I made 8 jars, two of each.  Both jars of the first three
types were contaminated by bacteria (slimy ring in jar, cloudy, etc).

So, while the Jalisco sat, I made eight new jars....even
going so far as to PC the shit out of the empty jars before starting
to make the LC setups.  ...again both jars of the three strains failed,
and the same way.

Now I have four jars of Jalisco and none of the other three.

So, I decided to inoculate petri dishes of each variety, so that I
can see for myself what it up.  I do seem to see the "coffee stain"
looking circle around the growing mycelia.  I will try to get a photo
up tonight.  I am pretty sure that either the syringes arrived contaminated
or I contaminated all three of them the first time that I touched them. 
This is odd as I have not had this problem with any of the previous syringes
that I have purchased. (GT, Roatan, PE6, Pan Cyan, Texan...) 
They are about two months old now...perhaps that is a factor.

LC's are unreliable.

Bacteria on agar is unmistakable. Post some pics and I'll give my opinion.

This is the Jalisco dish.  It is hard to see in the photo,
but it is very clean.


In a way these results are validating.  My LCs were failing
because they were doomed.

Will I have to deal with this every time I do not use
a syringe immediately? (or, well, within 3 months?)

Any idea appreciated.

You seem to have a lot of issues with contamination.  What is your sterile procedure?  I like to go MS to LC to agar.  LC colonizes the agar right away and can be cleaned from there.

Is that egg white pattern growth on the agar? Or just light tricks from the lid?

There's a risk with any syringe.
Bacteria are everywhere, it's very difficult to make a truly sterile product, and a spore sprint is never sterile.
So when a vendor makes a syringe from a print, there may be bacteria present.
It's not necessarily the case that a mistake has been made.

Generally, a low contamination rate will allow some jars to colonize thoroughly, and then you use those to propagate healthy mycelium.
