cloning question
    #13012943 - 08/06/10 03:04 PM (14 years, 5 months ago)

i've been looking around on cloning teks and found one that seems easy enough for me to venture down as a first time attempter. it's atwar's cardboard cloning tek. my question is, and i don't know if i'm searching it wrong, rather than putting the myc to an agar, am i able to mix that straight into pasteurized horse poo?
i'm assuming so as it seems to be nothing more than a kick ass version of a cake getting tossed into the mix.
also, as an after thought, what is the best way, if there is one, to store cloned myc?
thanks guys and gals for your help

The best way to store cloned mycelium is in a slant.

And you should always spawn to poo using grains.

i've actually had some pretty good success with spawning brf cakes to poo. i was just wondering if the clone myc would be easier being able to skip that whole process and also produce sweet guys for certain

right, it's so automatic making the cakes that i space on what the ingredients are. so then what's the best way to get full use out of the cloning process. i've only used the brf and ventured into birdseed once with zero-point-zero success. still researching the agar part of things and for now that seems a little intimidating for me.  i love the idea of maximizing the monsters and really want to be able to broaden my horizons with this hobby. sorry if these are elementary questions.

Lots of people have success cloning by putting the clone to LC.

From there you inoculate grains, whether its whole grain or BRF.

Then you can decide if you would like to spawn or fruit from cake.

Well when I clone I take a piece of tissue to agar petri dish.  Then once the dish colonized I take a piece of tissue to either a grain jar or an LC.  If I go to grain jar I let it colonize then take that grain jar and do a G2G with it turning that one jar into 10 jars.  Then once those colonize I use the 10 jars to make mono tubs.

If I use LC I let the LC colonize then suck some up into the syringe and knock up some grain jars with it..

I like going LC to agar then out from there.  I don't like how long it takes spores to germinate the agar media.  Clones always go to agar first.

I finally got to this point, and I wonder now why it seemed so hard. It's really not.

Cloning and agar work is simpler and more fool proof than LC.  Why do we give 2 shits about LC's, why is agar so intimidating to the beginner?

Before I got a flowhood agar was far out of my reach. My house is so loaded with spores if I look
at something funny it will have green mold. I don't know if its because of where I live, or if its
because this is the first house I've had that is forced air.

Even with the flow hood, my contam rate on agar is about 4%. I believe this will improve with my technique though.

it's intimidating to me because of the use of agar and a petri dish. those things sound very high tek to my fairly noob mind. but in the more i search and read, it's seems the terminology is scarier than the processes. but then again it took me a year or so to finally use horse poo because pasteurizing sounds like pretty gorilla dick process. more than just boiling at a certain temp for a set amount of time. i'm glad i asked this question and thank you guys for your responses. i already feel a little more confident in trying to phase out some of the more tedious processes.
thanks again everyone.

Oh agar is super easy lol. Just that my environment isn't suitable without a hood. I'm sure
a glove box would work, I just hate how confined they are.

Pouring agar in a glove box sucks.  When pouring agar in a glove box, I like to use a small mouthed quart jar instead of a whiskey bottle, it fits better in the glove box.  Back in 06, I used to pour pre mixed PDA into pint sized widemouthed jars in my kitchen on the stove.  I am not recommending you do this but it can be done.  I have a flowhood these days.  Coolest fuckin' thing I have ever owned.

I think the same way I don't know why more people don't use agar...  I mean it really aint that hard.  After my second try or so with agar I had my technique down and knew what I was doing and now it is super super easy..

LC's are a bad habit for me, like smoking cigarettes.  I was raised on LC's.  Now when I walk into the cook house I just go for the LME/DEX without even thinking about it.  I have since then broke that habit and now I reach for the potato flakes and powdered agar with out even thinking.  Agar is way less work intensive, too.

Yeah I just PC'd 2 bags and 4 little half pints. I'm gonna test some agar I've been working on.

I don't think its quite isolated yet, but I'm still developing an eye for sectors.
