I have recently returned to H2O2 use in my agar plates, due to a major contamination problem, and have found that a few species don't seem to like it at all, and never seem to get used to it.

P. mexicana
P. tampanensis
all Pans

Happy on peroxide:
P. cubensis
P. azurescens
P. cyanescens


Anybody else find they can or cannot use peroxide with certain species?

My Panellus stipticus took weeks to show any growth. It eventually colonised the plate but took quite a while.

how much h202 are you using?
mexicana and tampanensis grow quite slowly on peroxidated agar, but it does grow, covering a 10cm plate in 2-3 weeks.

How do you actually use Agar with H2O2? As far as I know, the H2O2 will kill the spores. So how do you inoculate the H2O2/Agar?
And is spraying and rubbing with Lysol enough to sterilize petri dishes or do you actually have to PC them?

I also noticed that Panaeolus cyanescens, P. mexicana, tampanensis & atlantis grow slowly on peroxidated agar. But they do grow!

To be perfectly honest, ive noticed no slowness or weaker growth when any mushroom is transfered to h202 agar. I used a high level of h202 too - 4mls per 250mls of MEA mix.
Ive had no problems with panaeolus Cyanescens, Trops, Subbalt., Psilocybe cubensis, psilocybe cyanescens, azurescens, bohemica, weilli, tampensis, aucklandii, subaeruginosa, eucalypta, agaricus brunnescens, hyphloma capnoides, shiitake, shemiji, and various oysters and reishi. All seemed uneffected by the addition of h202. SOmetimes pans can take a while to pcik up when transfere, but ive seen this with or without h202

Yepp! Every thing i grow on agar is on 4mls h202 to ever 250mls of MEZ - i got the measurements wrong originally, but it worked well for me - especially as most of what i work with are wild prints.

everyone just ignored this guy. not even worth a flame. haha, i love the advanced forum

to the question though. in the tek, it pretty much says, the less peroxide you can use, and still be effective, the better.

my opinion, i think it may also have to do with the maturity of the mycelium. eg if you start agar with young mycelium, it may die, or grow more slowly than mycelium that has almost shitted a flush, and is in its prime.

one other idea i had. i was thinking, for an experiment, maybe someone can develop a peroxide friendly strain. if you keep cultivating one strain with agar using peroxide, maybe later generations will generate the peroxide converting enzyme more abundantly. this can speed the process for the slower colonizing strains on peroxide. this idea stemmed from darwins philosophies. only the strongest survive to generate more strong offspring. also known as natural selection, and more commonly known as evolution.

I use H2O2 on all my agar plates and have had no problems with contams, furthermore I tried Rush Waynes " ten minute" grain spawn H2O2 treatment
Not only did i not get any contams it appeard to yield more
The strain I used was P cubensis EQ

The problem with doing trying to make h202 friendly isolates may be that the mycelia may become reliant on the h202 in the same way that PF classic relies on brf alone.
Funny, my latest bunch of panaeolus trops (probably incorrect identified and may have 4 spored basidia) ive been watching carefully and they have visibly slowed down when introduced to h202 agar, whereas the Weilli was fine.
Never noticed before, but I dont see it as a problem.

This would cause me to conclude azurescens grow slower on peroxidated agar. I believe the pic was taken w/o a flash because condensation would obscure the image otherwise. The two plates on the top right are azurescen isolates. All others are cube isolates except the left section on the bottom left plate, it is a multispore.



Of course it could be the medium as well, DFA.

I will add that my mexicana A clone from sclerotia is TAKINg a lifetime to grow on 3 ml per liter 3% peroxide.

First experience with peroxide and not liking it much for the mexicana A I got. Will try with some cubensis clones,before throwing it away forever.

slowing down the isolates can be useful - if you make up 10 plates and you only used one or 2 per week - or you get sidetracked you ca come back and know theyll only be slowly colonising and wont have reached the edge yet

Of course the best way to store them after this is sealed in mycosyringes where they can sit for months till u need them

funny, i was thinking the same thing, once I realised that the mexicana does adapt.  It leaves you with a three week window.

First two weeks almost nothing happens then bang the third week the mycelium takes off.

The cubensis just ignores the peroxide.

I have decided that I LIKE peroxide. :laugh:

Not wanting to bring back the dead, I just noticed Ps. baeocystis has not been mentioned here.

I used a small amount of spawned woodchips to inoculate a modified Rush Wayne preparation, oak pellets supplemented with 5% urea (based on weight of water added) containing 0.3% H2O2.

The mycelium grows quite slow under these conditions but appears to be doing fine.

I have used much higher concentrations than most of you. I used 0.3-0.5% H2O2 on Ps. cubensis "Thai Koh Samui", which did very well, on Ps. cyanescens with no real difference in growth rate noticed, and on Pan. cyanescens with also the notable slowing of growth as has been mentioned here.
