moisture and SDA plates

Some dude streaked SDA plates about a week ago with fairly old spores. No growth is apparent yet. Should he have added 0.2ml of sterile water? Will that hurt colonization?

If mixed right , agar contains all the moisture needed to get spores o germinate . Age and viabilty of spores may be an issue .
What temp is "dude" incubating this in ?
Yes adding water could bring on contams , just opening up that petri could be a mistake . So wait some more .

The dude innoculated in a laboratory with a loop and all the fixins. He used the quadrant streak method for isolated colonies. He asked his prof about the pH of the SDA and she claimed that it was ~4. He thought this was too acidic for P. cubensis, but decided to give it a try anyway since the facilities were accessable.

The plates are incubated at 27 deg C. For all you americans, that means about optimal colonization temp for P. cubs.

The plates were already supplemented with the extra water. Sterile water and a sterile pipetter was used, so contams should not be a factor.

All the stuff the dude needs is right there at school. Its too easy to not try.
