clone tissue not all touching agar
    #1277215 - 02/04/03 09:14 AM (13 years, 10 months ago)

Ok I cloned a nice mush on some agar plates. I broke th stem open and pulled strips of flesh from the center. Then I layed the strips on the agar. The prob is the strips are about 2" long and they only touch in the center. The ends curl
up a little. There is also a lot of blueing on the tissue, is that ok?
Next time I will use smaller pieces of mush tissue. Do you think that there will be any probs with these plates.

You only need a very small piece touching the agar, as long as the tissue still has living cells in it, it will take. All clones do not take, so it's hard to tell.

I may be wrong but I remember reading in either the tmc or another of stamets books that pushing the tissue down into the agar would provide the tissue with a better chance of developing. Its what I always do with clones and Ive had pretty good success.
