How much air does mycelium on agar need?

Hi!

I was wondering, how much air does mycelium actually need to grow? When you inoculate a petri dish of agar, and put some plastic foil over it, will there be enough air under the foil for the mycelium? Do you need to poke holes in the plastic foil or how do you go about it?

The dish with a lid it comes with is fine , no need to wrap in foil or plastic , if you have problems with agar drying out too fast store them in a tupperware container with lid partly sealing , leave one corner loose . Do not fan or open dish until ready to harvest strain , transfer etc .......... you will get contams if you open and fan . :tongue:

Oh, they come with lids? Didn't know that. I know the petri dishes used for chemitry, and I've never seen one with a lid. That's why I was asking... But I guess it'll be ok to use a dish without a lid and just put some plastic film around it as a protection?

Do self a favor before trying agar plates ....... buy some new petris , that are sterile (most are when new ) , plastic disposables are fine , glass is best but costly . Wrapping with plastic will result in lots of failure IMHO !
Sporeworks sells them for , I think 15$ for 60 of them .

I always seal them with a glad plastic wrap, where I cut a small piece of the roll.
This prevents both contaminates coming into the dish for whatever reasons, ads for easy handling since the both halves would otherwise come apart and it allows gas exchange as well, since the polyethylene wrap is gas permeable.

Is'nt that a pain in the ass when comes time to harvest issolations ? Having to remove film and all . For long term storage , I have considered wrapping with parafilm , but during strain issolations I like easy access . Just must handle with care (I only have one thumb on each hand ) so lid is no problem .
I assume this person has petri with no lids at all , hence the advice to buy some new dishes .

  anno, would you happen to know how much gas permeability polyethylene has? could one substitute, for example, polyethylene for polyfill? would that allow enough gas exchange for myc to coloniz a qt jar? :smile:

I'm the lazy type , but will try that to store them , saran wrap is cheaper than parafilm . Thanx for the idea ANNO ! Opps sorry about the misspell !

ps What did you use to make that nice clean cut on a roll of wrap ?

NOT saran, Glad Wrap. Saran is a different material, an it is not gas permeable.
