reviving old spore prints

Howdy,

My first post, but I've been reading the forums for a a month or so now.

I realise may have been answered already, but I want to start posting also and stop lurking. So no smackdowns please.

Just recently rediscovered my old grow stuff from 7 or so years ago and I'm looking to see if can get some of the spore prints to germinate. They were taken on glass, in a jar with the lid sealed tight.

I understand that this can be difficult with the spores being so old.

Anyway I've got some agar that I've just inoculated with multispore from a syringe made from the spore prints today.

So It'll be a while before I know if I've succeeded. In the meantime I've got a good supply of more petri dishes, are there any other tricks I can try in the agar/nutrients department or ratios to boost my chances of success.

How successful is LC for example from multispore of this age?

I'm in OZ so a little wary of ordering from overseas as I did read somewhere that syringes coming through the mail were getting snatched at customs, plus I don't want to fly any red flags if I can avoid it.

A few prints from overseas shouldn't raise any red flags, and there are a few Oz members here you may be able to persuade to send you prints in the mail.

As for your agar, it's standard practice to let the spores soak for at least 24 hours in the syringe prior to inoculation. With 7 year old spores you may want to wait even longer. I predict failure but you can always try again. I'm no expert by any means, just my opinion.

It's hard to spot contams in an LC.
Agar is your best bet. You may want to hydrate your 7yo spores for up to a week.

The previous poster's response seems cogent.  If you try agar, then
you should be able to isolate a strong looking sector (and repeat
to isolate one strain).  I am getting ahead of myself, as I have yet to do
this myself, but I have procured the materials to try.  Isolation
would be impossible using LC....except perhaps when used to inoculate
agar.

I have already made a few mistakes that isolation can cure.

The spores that I inoculated this batch of agar with had been re-hydrating for 36 hours, so I'll wait for a week then if nothing has appeared try another batch.

I will likely need another batch or two of Agar to isolate any contams then go to LC and through to bulk grain.

Hopefully something comes back to life, if not I'll start shopping for prints.

Forgot my manners also, so I'd just like to offer up a sincere thank you to everyone freely giving help and advice on this forum.
There's a lot of time and effort that's gone into sharing TEKs and information it's very much appreciated.

As said above, it's rather unlikely for seven year old spores to still be viable, but it has worked in the past. Even older ones. It really depends on the spores and the way to storage.

Agar would indeed be your best bet, since this way you don't have to waste any substrate materials for probably non viable spores.

You just have to try it.
