air exchange?
    #1261221 - 01/30/03 09:50 AM (14 years, 1 month ago)

I have seen posted here before suggestions to give incubating jars some air exchange by untaping a couple of inoculation holes. I have usually seen this suggested when jars are at about 70% colonization.

Just to clarify, is that estimated 70% total colonization, or just 70% coverage of the outside of the cake?

I ask because my jars have all been doing pretty well, but some of them seem to have 'stalled'. They seem to have plenty of moisture so I don't think that's it. Basically, one has been birthed already and three more will be birthed this time next week, but the rest all have unexposed patches on the bottom that never seem to get any smaller. I turned them over but that doesn't seem to have helped. I'm thinking of untaping some inoculation holes on the other jars but I'm not sure if they're far along enough to resist potential contams from that yet.

the reason for turning jars upsidedown is to let heaver then air co2 out .if the jars are sealed tight this wont happen .

i usually untape all four holes on  my jars about a week after the mycelium starts to spread. by this time i can spot any contamination, so it's safe to uncover the holes. the whole purpose of the top vermiculite layer is to protect your subtrate from contamination. don't be afraid to try this. i have never gotten a contamination AFTER untaping the holes. then another week or so later i turn my jars upside down. by this time i usually have 75%+  colonization.

the top part of your jars colonize first, and once the mycelium has reached all the way around the jar, it's pretty safe to untape holes or turn the jars upside down. this is just my opinion, but i feel it's much harder for contaminants to reach the uncolonized substrate, because they have to get past  a "ring" of colonized substrate first. i hope this helps.

as always, please feel free to correct me if i'm wrong about any this.

I do not cover my holes back up after innoculation. The layer of pure vermiculite on top of the mix will keep contams out. This seems to work well

me neither. i cover with foil to sterilise, take off the foil when i inoculate, spray the outside of the jars with antibacterial spray, then stick 'em straight into the incubator.

yes, all that is true. there's really no need to cover the holes after inoculation. i just do it because i'm paranoid. during the first week after inoculation you don't really need air exchange, because there's no mycelium yet. i always remove the tape after i see mycelium though.

I'm the same as you. I like to wait till there is some myc growing. Even then I'm paranoid so I use a idea that Skikid16 gave me. Coffee filters and rubber bands to cover the top when I take the tape off.
