Pf tek colonization question
    #12599754 - 05/20/10 05:46 PM (14 years, 7 months ago)

hello, I've recently started three batches of Pf Tek jars-- my first, second, and third tries at cultivation.  In my first batch, I put a little bit too thick of a dry verm layer at the top and had trouble getting through it with my syringe for inoculation.  I corrected this in the next 2 batches, which are coming along beautifully now, but this first batch is beginning to grow mycelium at the top of the substrate instead of the bottom, just below the dry verm layer.  I'm going to leave them for now, as it doesn't smell or look like there are any contaminants present, but I was curious how colonization beginning at the top of the jar would affect overall colonization, and if having some of the spores exposed to the dry verm during inoculation would increase my chances of contamination.  Any helpful comments from someone who has experience would be much appriciated, as I'm very new at this and very curious about all possible variables in the colonization process.  Thank you

did you use 4 holes on the outer part of the lid, then make sure to push the needle down along the edge of the glass so you could inject your spore solution on the outside of the cake. or did you just inoculate in the center? it really shouldnt matter, one way or another its going to colonize the whole cake if you did everything correctly.

Yeah i used 4 holes, but didn't quite put enough substrate in on that first batch, so the syringe didn't get all the way through the dry verm on all the needle holes.  I already had everything sterilized in my glovebox, and the needle in the first hole before i realized what the problem was( I know I should have checked that earlier, but stupid beginner's mistake) so I elected to continue on and correct it next time. In most of the holes the syringe got to the top of the substrate, but some didn't make it all the way through the dry layer.  Regardless  of whether it made it to substrate debt, I still pushed it to the side of the jar and inoculated 7 from one syringe. Very few of the inoculations visibly rolled to the bottom of the jar, like my newer batches have.

Yea if you injected the spores into your dry verm layer then that sucked it all up and those spores will not colonize. As for getting contams because of that its possible, but only if the syringe wasnt sterilized or you sucked in unclean air. Gotta inject down the sides of the jars so the solution will run down, that may be the reason why only the top is colonizing. Another possibility the bottom isnt colonizing is you burnt it during pc'n or perhaps the substrate at the bottom wasnt left lose and you compacted it more, im not sure exactly what you did though.

Out of those things, I probably just injected the dry verm with a few of the holes and lost a lot, but all jars show evidence of beginning colonization at the top of the jar and its making progress.  Is there anyway I can help encourage it to colonize downward more?

Less you mess with it, the better off you'll be.
