I personally like this one and adaptations like it that add gypsum, coffee and/or manure.

You can but with a monotub you'll have better luck using colonized grains to spawn to a bulk substrate like hpoo/straw or coir/verm, etc.

No.You use the grains (WBS,Rye,Popcorn) as spawn. The bulk substrate can consist of many things but with whats listed it's pretty much coco coir/vermiculite mixture.
the spawn to bulk ratio is ok anywhere from 1:1 to 1:4,but the higher the spawn to bulk ratio the faster the spawn will colonize the substrate and the less chance of contams taking a hold.Using an 18 gallon tub and damion5050's coir tek you should get 10 quarts of substrate and use 5 quarts of spawn for a 1:2 ratio.
Damion5050's tek uses an 18 gallon tub and calls for 1 brick of coir,2 cups of vermiculite and 4 to 4.5 quarts of boiling water (try to keep the water at 160-180 degrees)....just read the tek.
