Bulk Liquid Culture

I would like to create massive liquid cultures (say in 5 gallon water cooler drums, for example). The problem is that the containers I want to use would be too big to fit in a pressure cooker. Based on Mycelio's findings in this thread, I think that the following procedure might just work to allow for the creation of liquid cultures without sterilization. I'm posting it here first to see if anyone can find any obvious pitfalls before I try it out.

PROCEDURE:

1. Around 100g of rye berries are soaked in water for 24 hours, then drained, rinsed, and placed in a loosely covered mason jar containing enough filtered water to completely cover the grain.

2. The jar is left in a warm environment for 3-5 days to allow for fermentation by the various lactobacillus native to the grains. Any grains which float to the surface of the water are removed.

3. The contents of the jar is strained through a clean cloth. The filtrate is collected and grains are discarded. The resulting liquid is stored in a refrigerator.

4. Several 5 gallon water drums are filled with a solution of distilled water containing 4g/L potato infusion solids and 20g/L dextrose.

5. Each drum is inoculated with a portion of the lactobacillus culture created in step 1-3, then sealed with a Tyvek filter and left to incubate for several days.

6. A flame-sterilized needle is used to inject a previous liquid culture (made via one of the many small-scale teks) through the Tyvek on each drum. The mycelium feed on both the remaining sugars in the potato dextrose broth as well as the bacteria which culture the solution.

It would be much better doing them in separate quart jars that way if one infects you dont lose everything

Agreed.

I do believe that the experiment on a smaller scale would be very interesting and even beneficial, however using it as a means of production,
especially on such a large scale, seems impractical.

I would like to see you use that procedure in smaller jars, just to see if it works.

Then if it does you could slowly increase the volume if you find that it is an efficient method.

Could you use metal drums instead of plastic and just boil the crap out of them, then inject with a generous amount of GLC?

Use gallon glass jars.
Agar/petri slice to LC.
Metal containers & boiling doesn't cut it.
You need to PC an LC, to insure its sterile.
