Transferring WBS jars to ziploc bags?

Ok so these WBS jars (made from g2g transfer) are at about 25% but only colonizing at the top and slowing down. They were filled a bit too full, so they don't really have enough room to shake and spread out the myc. Would it work to transfer the jars into big gallon ziploc bags inside a glovebox, so I could break them up and spread them out. Then I guess transfer them back to the jars so they can keep getting GE? Anybody see a problem with this idea or a big source of contams?

try it! if your in sterile conditions i don't see how it could mess it up as long as you make sure everything isn't just spread throughout the bag

I would highly suggest you don't unless you were using a flowhood. From my experience uncolonized grain is difficult to work with in a glove box without introducing a pretty high contam rate. I'd just try to shake them as hard as possible and if that doesn't work just let them colonize as if they were a brf cake and then break them up at 100% colonization either by hand or in a bag.

Hmmm well I went ahead and did it before the 2nd reply, so I guess I'll just have to wait and see how it turns out. The G2G transfer was originally done in the same glovebox, same sterile procedure and had no contams with 20 jars, so hopefully it will be OK.

Yea man you'll probably be all right. When i first started i could practically do g2g in open air with no problem now I don't even attempt it though cause my mold spore counts a so high last time I tried it every single jar contamed. You'll probably be fine I just normally don't like to take chances if you don't have to, rather wait a extra week then grow mold.
I actually think the technique of using ziploc bags to do it though is a good idee. May want to put some alcohol in the bag though never know if ziplocs come sterile.
