poking holes in lids
    #1241693 - 01/23/03 05:26 AM (15 years, 5 months ago)

i have read from different sources that you can poke one hole in the middle and try to reach the glass with your syringe. or you can poke 4 holes and reach the glass.....
Now that i've gotten lazyer i would like to try the one hole method, and was wondering if it really matters where you squirt the spores?
i know it's benifitial for the spores to run down the glass but what if it's just dumped in the substrate, how much difference would it make? (timeing wise?)

Fewer points of inoculation = longer colonization time.

i used one hole before but made several inoculations, rotating the needle in the jar each time.

Longer colonization for sure. If you're still prepping lids, try doing a whole shitload at once and get it over with. I have had some success using the tall thin half-pint jars with one hole, but I squirted the same amount of syringe water in the hole I use in the 4 hole method. Also, it had to be done slowly for good results. So, it didn't really save any time.
