Ive made a couple of syringes about 2 weeks ago now, and Ive inoculated some rye jars 4 days ago, and at 28 celcius, still no signs of germination.
Could this be normal? the syringes were made a week prior to inoculation.
But I noticed on one syring that the spres had made almost a cobweb like structure, almost like I'd spat in the syringe and shaked it up!
The procedure for the syring was the following...
Make print from nicely open cap in a sterile jar.
Wait a day for the spores to drop then take out cap and wait 2 days for the spores to dehyrate ( I had a lid on the jar with a polyfill plug.
Boil water put syringes in and squirt water in and out a few times.
Keep the water in the last time, and when cool (the day after) scraped the spores from the glass with a sterile rod, through another hole in the lid, then shoot water in the jar (through the hole) and suck it back up.
The solution had afew clumps but was not overly crowded by spores.
a week later there was (as I mentioned earlier) a a strandy  structure inside. I inoculated anyways.
these still maybe germinate in the next few days but I doubt that. :uhoh:

It can take 14 days to show signs of germination after inoculation.

blimey if they dont germinate that will be a waste of electricity!

Your 'strandy structure' sounds like it might be mycelium.  What color is it?  It's possible your spores have been germinating in the syringe.

sort of greyish, and Ive heard that pre germinated spores show myc even faster in the jars than if they werent already germinated.

Yes, if they're already germinated - sounds like maybe yes - and assuming you got some of the myc to come out when you inoculated, then it should probably show growth more quickly than just spores.  Still, it's probably too soon to call it a fail. :smile:

last night saw some white myc spots! never waited that long if I remember rightly. That made my day!:smile:

I inoculated to some rye and some showed growth in12, 14 days some others are just now showing growth 28 days later. Patience is key! I agree w/ andymc about the snotty appearance. I've gotten syringes in the past that totally looked like a loogey and they were just fine.

some jars have hardly any signs of growth. I think its probably better to inoculate non germinated spores because they will spread more evenly throughout the rye and each individual jar. if one end of the strand of germinated spores goes through the needle, the rest probably gets pulled through aswell. all into the same jar if you see what I mean.
Its weird though, I thought that homemade syringes should germinate faster since the spores are fresher and there are more of them.

If spores germinate in a syringe.
They often clump togather, in a stringy mass.
Often, more myc goes in some jars, far less in others, sometimes none.

Same thing for LC's

so extra shaking of the syringe would help. Or is there a way to keep the spores from germinating inside the syringe in the first place? I know normally you put in a chemical that stops them clumping (cant remember what its called) so does that chemical stop germination too? should I store syringes in the fridge? and if the spores have started to germinate inside the syringe (even if not noticeable to the eye) will refrigerating them do any harm?

Shaking helps break up spore clumps, as well as myc.
I store syringes in a cool dry clean place, in a sealed bag.
Fridge works to, but are not all that clean.
Usualy spores don't germinate in a syringe.
If the water was sterile.
But, it does happen
