Nice write up, very comprehensive - doens't leave much room for contaminants. which is always a good thing.

I hope people aren't scared off though, as you can happily do spore prints with much less attention to detail and still be ok.

My first times printing were on a bookshelf on a tea towel with a drinking glass covering half the peice of foil over night, and they always worked fine to start liquid cultures with no contaminants (open air inoculation). I did sanitised the foil first with alcohol wipes before printing, and then once more on the air exposed side of the foil (and allowing it to dry) just before folding up the print.

This worked well for a few months before I started to pressure cook the foil strips and print in a sanitized container with a filtered lid (drilled holes covered with micropore) -



This is now the preferred method as you just take the caps out and allow the prints to dry for as long as you need before folding them. I think this is as sterile as I'll ever go unless I start getting conamination problems. It needs to be said I live in a very dry arid area so this helps when warding off contamination problems... humid areas would be a very different story.

Yes I believe so! Just be careful and sensible, but pressure cooking your strips in a normal grain jar with a filtered lid for 20-30 minutes, or oven baking them as described in this tek, would be preferable.

Just stumbled on this thread, know it's a little old but great job. You make me feel so lazy! My prints are OK for sure but any way to clean up agar work is good news to me. Will try this tek and let you know if my contam rates decline. :thumbup:

Really good guide, I like the perfectionism and techniques you use, for simple stuff, such as the knife tech you used when folding the foil in order to seal the prints. Appreciate it!
