a couple questions about Tyvek
    #1217364 - 01/14/03 11:02 AM (14 years, 7 months ago)

1) How big should the main air exhange hole be.. (using 1/2 pint jars, 2 peice metal lids)
2) Ive read a few people people prefer to place the Tyvek on top of the metal lid, then screw the ring on. Is this just personal preference or does it actually perform better?
3) Let me know if this sounds right.. 1 main air exhange hole in the center, 1 smaller innoculation hole near the rim. Put substrate into jar. Either place tyvek on first or second(depending on what you all reccomend). Screw metal ring on. Wrap in tinfoil. Steam the the substrate. Let cool. Remove the tin foil. Innoculate through the small hole. Cover innocualtion hole with tape. Then store in a dark warm place until fully colonized.

Any other tips are appreciated. First grow and could just use some a little advice  I have been reading over these boards for a few months now and i feel comfortable to try for my first time.. ive got all the materials but just need a thumbs up before i start.

The way I do isn't the only way, but...
I drill one small hole in the lid, size doesn't really matter.
I put the lid on underneath the tyvek filter, then screw the metal band on, then wrap in foil. Then I pc and let cool. When ready to innnoculate use a flashlight, or other light source to find the hole. Use a second piece of tyvek to cover the top. Works good.
I like having the lid under the tyvek, just cause when you shake the grain they can be kinda sticky and may stick to the tyvek.

a 1/2 inch hole is plenty big enough. You just want to be able to get a little air exchange and be able to fit the syringe through.

i was planning on doing 2 holes.. 1 for air exchange(in the center) and one for the syringe(near the side, which i would cover with tape after innoculation). this way i wouldnt have to take the lid off to cover the hole in the tyvek with either another tyvek filter or to cover it with tape.. i could just tape up the small hole on the metal lid and leave the other hole how it is.

If you do it that way put the lid on after the tyvek. I put the tyvek on after the lid. I add a second layer of tyvek after I innoc. Both will work.

I just re-use the lids I made for my BRF 1 pt jars with the four holes. Then I put the tyvek on top cuz it looks cool and clean and you can write on it. Seems to be working good on my birdseed jars.

Do 1 QT millet jars this way. Spray Lysol everywhere then wipe everything down. Now that we are clean fill jars with 250 ml of millet and 130 ml water. Then I just cut my Tyvek about 1 inch to big on all sides lay 2 sheets on top of jar then lid on top of the Tyvek press down hard and tighten snugly. Then PC for 1hr at 15 psi let cool then shake the snot out of them to help prevent clumping and to help evenly distribute the moisture. Then remove the metal lid with out removing the Tyvek then put the ring back on tight. During this process you should be as sterile as possible spray Lysol before you start. Then be sure to have a clean needle flame sterilize it then wipe it with alcohol one hole in the center of the Tyvek is all you need inject with about 3cc of spore solution then tape over the hole place in incubator and wait.. This way you have maximum air exchange and real fast colonization.

I your situation I would do like above and just put tape on the 4 holes where you inoculated. Use 4 holes no matter what and shoot the solution left and right from each hole with the needle against the glass it will help speed things up dramatically. Are you using a verm seal? I would ad least do one jar this way to see. I don?t see why it would not work. but their is no need to have metal lid to unless your worried about it dying out.
