Semperviva germination times?
    #12158889 - 03/07/10 12:33 PM (13 years, 10 months ago)

I've been reading alot about this species but I'm seeing different information on MS germination times.
I inoculated some standard PF cakes with a MS syringe I made from a spore print (spore print was several months old) on 2/27, it is 8 days to the day and there is no growth in jar at all. They have been incubating around 80F. So being unfamiliar with this species, was just wondering if this is normal?

thanks
MM

Germination time depends on many factors. If the spores are very dehydrated it will take longer for them to germinate.

I've had Ganoderma spores take 2 weeks to germinate on agar while spore from the same print took only 4 days to germinate on another plate. I've had morel spores germinate in under 12 hours.

Give it another week or so. I'm currently having the same problem with some Lepista nuda spores that are only 4 months old.

Thanks for the quick reply BG!

Ya since the spore print was pretty old, I assume it was pretty dehydrated. I have some on agar too that show nothing. I was under the impression that this was a fast grower from what I read but I think the age of the print is going to be a factor. So looks like this one will just be a waiting game, but hopefully will see something in the jars in another week.

I have P. Semperviva on agar and also inoculated RGS with it.
I've had it germinate on agar in 3 days,
but colonizing the grass seed is painfully
slow, although my temps are a little low (mid 60s).
AFAIK it's a pretty slow colonizer and slow fruiter. Fruit bodies need 3-4 weeks to fully mature in a very higy humidity conditions.
Good luck with your grow:thumbup:

Thanks for the replies.

Well still no germination on agar yet and no germination in PF jars yet either. Was inoculated on 2/27 and the print was from last summer. Incubating temps are 79-83F. Hoping to see something soon though. Will update this thread as I go along.

Do you have a microscope?

In the past I had a few print of various species that refused to germinate and when I put them under the scope, most of the spores were cracked or busted open. (aka dead spores).

The Lepista nuda spores that I posted earlier about, still haven't germinated on agar for me. Its been about 16 days now on agar.

I don't have a scope anymore unfortunately, used to before I got into mycology.
That's very interesting on the busted spores though.

When I made the syringe I only scraped about 25% to 30% of the print so I could save some for later. And the print was a rather small print to begin with, probably no more than an inch diameter. No spores were visible to the naked eye once syringe was made, but I know that sometimes that really doesn't matter. Also the print was kind of old, well, old as in last July. In three more days it will have been two weeks since inoculation. Anyways, just throwing a few more details out there. I think I'm going to scrape more of the print and inoculate some more jars today or tomorrow.

One thing I didn't consider was the print came from Europe to USA and may have been irradiated. Any thoughts on this?

Oh and the print was from 3/09 not 7/09 which I stated earlier.

Well 2 weeks and no sign of life, hmm I might just chalk this one up to a failure or my just wait some more to see if anything happens.

Have you tried scraping dried spores directly onto agar?

Hi BG
Ya I have some on 3 plates, that was scraped on 2/27 as well. I took out the plates the other day to look at and it does look like there is something inside one of them. Little white dots about the size of a pin dot. Will have to get some pics up soon. A couple days ago I went ahead and used the whole print for a syringe, now just need some more BRF to inoculate or might use RGS.

Well just to update, no germination in those jars, so pitched them.
Not sure what the cause was, maybe not using enough of the print.
So I got another print from same person, although its the same age of last print and inoculated some PF jars. I used the whole print this time and noticed germination in 3 days already!
