When transferring spores to agar.

Should i just put just spores on the agar with the innoc loop or should i add a drop of water with them? I have just some tiny prints that i want to transfer to agar to keep them going and don't want to mess them up. thanks. have to head to work but appreciate all responses.

I think the usual method is to flame the loop, run it through the agar to cool it, then pick up spores with it.
I don't think you need to add liquid water, there's plenty in the agar medium.

i still got nothing on my agar that i transferred from spores to agar on the 5th on feb.

2 more questions:

1. can i just put some of these spores into a lc?

2. also, i got a few syringes of some edibles that i want to make go further without making 3 more lc's from them. Can i put some of the lc onto agar and will it grow out? or will there be too much liquid and not enough myc for it to grow.

You can use a few drops of LC and put it on agar. Spores in lc is also doable.

Let the mycelium grow out and transfer a piece of wedge to an LC.

Yes, that way any nasties left on the spores will be long gone if you take a piece of mycelium from the outer most edge. I recommend three tranfers that way you can transfer rhizo mycelium to the LC.

I put 5 cc's of ATL #7 lc into 500 ml of lc media today

and some spores from a jalisco print into an lc media today. they're both on stir plates atm.

couple q's again.

1. when i put the spores from the jalisco print into the lc i couldnt get any off with the innoc loop so i had to dip the loop into the lc then onto the print and back into the lc a few times. (the loop was sterlized of course) does this sound safe? having to dip it?

2. also, about how long will it take on average for the jalisco (spores) lc to start to see life?

yes, the jalisco at least. it looks like the ATl is starting to get a foothold on the agar but nothing from the jalisco.

ive never really had any luck with spores on agar anyways.

also somehow a damn dog hair got into my jalisco lc while i was transferring spores into it. i dont know how that happened, i was in a glove box with sleevs, gloves, etc. :ohwell: i dont have much hope for the jalisco anymore.

anyways: how long before i see life with spores typically?

Did some cyan and it took about seven days. I woke up one morning and there were little white fuzzy dots where each individual spore had germinated. it was cool but felt like it took for ever. I made a weak spore solution and did one drop on the agar. myk

Spores to anything takes a while. I woulda made a syringe with the print and put a drop on agar.


LC on agar is a matter of days.

:thumbup: you might see some spore clumps germinating and getting fuzzy, then the mucous-like look of the mycelium. it could take anywhere from 1-2 weeks for it to get popping.

did you run a stir plate or stir the lc at all? I have mine stirring about 20 hours a day on low.
