I would like to know what a ramsey seal is? I also would just like to tell you how I go about innoculations. Fist I do all of my sterile work in a small closet (don't have a walk in unfortunatly) my first step is to spray down all of the walls and floor with 10% bleach water then wipe down. Then I turn off my hepa filter and spray a mist of bleach water into the air and close the door and take a shower in the mean time. Then I put on clean clothes and use a hand sanitizer on my hands and forearms then work quickly and effeciently. If that doesn't work for you then its time for a laminar flow hood (my next investment) I hope that this could be some help to you.

jonah I dont think that a glove box is necessary when using the ramsey seal sounds to me like you have just had some bad luck. Make shure that you pressure cook long enough and when you inoculate make shure you sterilize the needle between inoculations also don't forget to turn off all fans and Air conditioning and don't breath on your jars and cover them with tape when you are done. I think thats it. Good Luck

I think some people over do it... you dont have to be as sterile as you think, I would think that just flame sterlizing the needle, popping it in the hole (breaking the tap) and putting in the spores, then quickly recovering the hole with a little more tape, this way you are not fiddling around doing one thing and letting the bad spores get in while doing this...

phrozendata is right, that all seems pretty extreme for sryinge innoc...only jars I ever lost were due to condensation forming on the inside of the lid during sterilization, wetting the verm layer...ramsey seal solved this, no problems since...all I do is wash hands before & wipe needle w/ alchohol between each hole...btw if you do flame the needle, be sure it is totaly cooled before innoc, or the heat will kill the spores, probably the reason for zero germination...

When using the oven tek make sure the spore print is not set inside the oven or on the rack in the path of direct heat for very long. Heat kills fast.
