master jars transfer suggestion?

I have 2 quart jars of winter wheat berries, 100% colonized with amazonians, ready to go. I'm not sure if I should go the safer route and do G2G transfer or if I should try a LC.. these were multispore inoculated directly. also if I do G2G, would going another transfer from the 20 jars that i get from the master cultures be too much and possibly be pushing it, as far as contams and fruiting ability?

It should all be the same original DNA and thus not a different generation, so you could potentially take one of the 20 you get and turn it into a bunch of new ones, but don't waste all 20 on transferring to new ones, because then you have to wait again on seeing anything....as long as the grains are 100% and consolidated each time you transfer though, then contam shouldn't be in your masters, it's just the open air to the new jars that you have to worry about getting bugs. If you're using a laminar or glovebox and sterile habits(gloves, mask, alcohol) you should be fine during transfers.
I'd get a bottle of the spray alcohol and peroxide though, it makes cleaning on the fly really easy and makes sure you get every inch you spray clean.

i whip everything down with alcohol and spray oust in the air to kill bacteria. so should i just use one of the 20 to keep the jars rolling? at which point does the culture start to senencese?

Look up agars grain LC.. It's basically just taking all the myc out of your grain and putting it in some water, thus making an lc without the question whether or not it is contaminated.

Just use 1 or 2 of your secondary transfer(doesn't count as a generation, so there's no degradation of the strain) and transfer to new jars. Just make sure you don't have any excess water on the outside of your new grains, that's just asking for contams, but yes you could also use the Grain LC to hit them up, but I find the myc gets going a hell of a lot faster from a transfer than from brand new and unestablished myc.
