questions on mycelium isolation
    #12034186 - 02/15/10 02:07 PM (14 years, 10 months ago)

ive got a couple brf tek jars going and one in particular has maybe 10 different spore germination spots spanning from very rizomorphic mycelium (ropey) to very white thick mycelium that looks like extra thick cotton balls.

I plan to open it in a sterile environment and isolate a few of the different mycelium and possibly put it on agar (havent decided yet).

my question is this:
what are the fruiting effects of the different types of mycelium? ive heard rizomorphic is the type you want and grows faster but what do the super thick fuzzy ones do when fruiting? id like to extract a range of them and experiment, and im just wondering what to expect

(yes i realize this isnt the best way to extract the mycelium for agar, im just experimenting a bit on my second grow)

The only problem I see here is that even if you are able to successfully isolate the mycelium without introducing any contams odds are your going to have multiple strains within each of the spots of mycelium you isolate there wont be just a single strain.

As far as what "kind of mycelium" will produce what kind of fruiting characteristics there's no way to say because it will vary based on your genetics. The only way to really know how an isolate will fruit is by fruiting it.

Agar work is an absolute must with strain isolation. Buy about 250 disposable petri dishes*.  That will allow you to test about 25 isolations. It will take several transfers to get an isolation. It would also be better to fruit your brf cake, and take a fresh spore print for isolation work. Your results will better if you go directly from spores to agar. Good luck.

*less if you buy the 3-way partition kind

I wouldn't say it's an absolute must. I've successfully isolated strains without agar but if you want to do it with much of a rate of success, yea agar is necessary. Otherwise it's a much longer process and your gonna end up with mold or senescence kicking in.
