Hi all,
Who can tell me if it is possible to add some Gentamicin sulfate to the masterslant tubes to prevent bacteria ? Or is it a bad idea to add Gentamicin because it's not good for the mycelium when you will keep it good for a long time ?
I hope somebody has information about using Gentamicin sulfate I make my masterslants on the same way as RR tel in his DVD "Let's Grow Mushrooms"

Leave the gentamicin for isolating, once you have health mycelium store it so that it does not get contaminated. Good sterile techniques will be of more use than anti-biotics.

I've had poor results using antibiotic agar in slants. It seems to have an effect on the mycelium when it's stored long term on it. RR

Hi,
Thank you for these info on the moment I use only gentamicin in petri dishes of wild species.
Is there any difference between MYPA and PDA agar what is beter for master slant cultures ??

A little off topic: Does anyone know if liquid media supplemented with 5-10% DMSO would work for storage at -70C, or -200C?

Doubtfull. You'd want to add the DMSO to skim milk and/or glycerin. You don't want to try and store it in actual media regardless of what you put in it.

You want to reduce respiration during storage so you want to use a storage media with very little carbon source (sugar).
You can look up storage techniques, just use the search engine.
Here's an example... http://www.ca.uky.edu/agcollege/plantpathology/vaillanc/storag.htm
You can store fungi with skim milk or glycerin as a cryoprotectant. There's other methods out there also, such as overylaying with mineral oil, etc..
RR uses a method using a small stick of partly colonized wood.
