Need help on mexicana sclerotia invitro

Sorry if this has been asked before but I'm still a relative growing newbie. and this is not in the faq. The recipe for the substrate is, but there is little other info on this topic at all. The search function didn't yeild much either.

Basically I want to use the correct size jars to get the most sclerotia I can. I was thinking wide mouth quart jars. And with the rye seed to be able to shake it it would only be partly filled. How filled should it be?

Another quick question, is it possible to sterilize grass seed in the microwave in a jar, my 4 quart pressure cooker probably wouldn't fit the big jars if those are what I use.

ANd when I get to the point of finally innoculating the jars how much spore solution should I use for each jar? I assume that when making shakable jars you don't do multiple innoc points like on a PF cake???

i'd use bags. you will have to fractionally sterilise large jars=hassle. microwave wont work well, dont even bother trying. 1cc of spore solution is enough. jars should be 1/2 or 2/3 full to allow shaking, remember the seed/grain will grow/expand when boiled

I'm not going with bags. Just to clarify you are thinking oven bags, right? It's jars all the way for me. I'de be concerned with ripping a bag and I doubt whether a bag could really even be shaken, and I don't think they're reusable either. I'll use the fractional sterilization you mentioned or buy a bigger pressure cooker if I have to.

I'm glad 1 cc is enough, that way 1 syringe will do a bunch of jars.

I am pretty sure he meant 'spawn bags' which have a filter patch attached for clean air exchange and can be autoclaved. They are very strong, large, and can take a lot of abuse (and be reused several times). Jars or bags, either will work.

With larger jars, you might be better off doing a spawn transfer rather than spore innoculation, if you have contamination problems. With more starting material already in its grow phase, contamination has less time and space to move into.

Why would you have to fractionally sterilise large jars? That doesn't make any sense. Sure when not using a pressure/cooker autoclave this is an option (and a lot of hassle) but there's no difference between bags and jars in this respect.

Both can be most easily sterilised in a pressure cooker (2 hours for 3 liter bags)

The grass seed/water mixture does not expand so much during sterilisation as rye or millet.

Spawn bags reusable? You are kidding me right? They are perfect for one time use but do you actually clean them out and use them again?

They are not very strong and can not take a lot of abuse. Sure they are strong enough to work with but they can be damaged quite easily when not handeled properly. Small holes will lead to contamination. For less experienced people i would not recommend starting with bags, start with jars and once you get the hang of that move up to working with bags.

Working with bags is a delight compared to working with jars but it has its own advantages and disadvantages.

I don't even know how to make a spawn bag. Do you just buy some filters and sew them in to an oven bag? I have no clue about that.

I'm not worried at all about contamination problems. I haven't had any in several months of doing PF jars. I use rubbing alchol and Lysol liberally before I start the sterile work and I have a glove box I made with a clear rubbermaid tub with holes cut in for my arms. I always sterilize the syringe with alcohol before each innoc injection as well, I used to use a lighter but the spore water would squirt out of the needle.

I actually do reuse them, becuase I don?t seal them with a sealer, but fold the top together and tape it.

They can be reused several tumes(at least 5 times) if you don?t mind the work of rinsing them with water.

yep i did mean autoclavable filter patch bags. i have reused some once or twice at most. they are cheap and i have 100s so i dont bother unless they come out in good condition

in the original post he said the jars dont fit in his PC. i am against him fractionally sterilizing, just pointing out that he would have to go that cumbersome way if he still wanted to use them

My friend is thinking about doing a sclerotia grow.

He is planning on doing just 2 pint jars. 150 ml grass seed with 75 ml water in each. Pressure cooked and multispore injection.

I have to use a polyfill filter right? I read this but just wanted to make sure(noob). Harvest after 3 months? Sounds like a good idea?
