Psilocin is structurally similar to serotonin.
How do you know which is the best? Do you take various drugs that affect different neurotransmitters and see which you like most? It's not very scientific, but it's a decent anecdote.
I've always been a fan of acetylcholine. I can't move a muscle in my body without it. Cannabinoids are always fun, too.

Well putting it shortly:
I like:
Opium, Hash, (not green leaf, I don't like it much), Shrooms, LSD.
Speed *ever now and then but not since we've had kids, can't be all out on stuff all night!, Xanax/Alprazolam
Since we've had kids, we mostly only trip every couple months or have a few drinks.

I believe classic psychedelics bind to certain serotonin receptors (ht2a, mostly), which causes the associated neurons to be significantly more or less likely to fire, among other less well understood changes. I'm no neuroscientist though, and I'm sure someone else can give a much more detailed and better explanation.
