I HATE CONTAM'S!!

ok i was just wonder wut peoples success rates are.  my friend has colonized (or should say tried) about 18 1/2 pint  jars over a period of time.  he has a glove box, sterlizes everything twenty times over and still seems to get massive amounts of contams.  out of the 18 jars i would say  8-9 were contaminated with either cobweb, green, or some kinda oranage/ yellow one.  now, he flame sterilizes his needles, and wipes with alcoho, as he always hasl.  is gettin contams unvoidable???  should my friends failure rate be over 50%, hows everyones else contam rate?  i guess it would make him feel better to know others keep failing misserably with tryin so hard not too.....  :frown:  (my friend just thinks he lost 5 more jars..... to some kinda green mold in his jars!, which by the way has no idea how got there.  syringe was left IN BAG in came in with put directly into glove box, then opened and sterilized the inc.)  to do a final test i inc. some honey water.  any help or suggestions would help, also if you could post your fail/success rates, just an estimate is fine if you don't know.

Where are you guys doing all this work?
Is it always in the same area? Maybe there is something in the air around that spot which is just happening to find a nice little spot to grow

Are you keeping a close eye on humidty and where your storing all of these? How closely are you watching them after you've inoculated the jars?

Sometimes the contam isn't from the spores or syringe but it comes in afterwords.

Just don't lose hope and keep trying

Does your friend use a pressure cooker?
What is the general cleanliness of the work area??
Homemade syringes??

Sounds like an above average contam rate, lets try to pinpoint the problem :smile:

Where does your friend do the work?  On the floor, in the basement, kitchen, etc? Does your friend shower before working?  Wear gloves while working?  Wear short sleeve shirts while working?  Does your friend flame the needle between each jar?  Does your friend open the jars before they are 100% colonized.  How much does your friend handle the jars?  Does your friend clean the incubator between runs?  The more info we get, the more likely we can help isolate the problem.  :smile:

aight, my friend has a glove box that is "air tight" two holes for your hands, a hole cut into the side of the box with a filter attached and a little fan is placed on the inside to suck in clean air through the filter while the air inside is blown out of the holes for your hands. clear lid on top. cleans hands. wears gloves. before using the glove box its wiped down with alcohol, then sprayed with lysol. then everything he will be using is wiped down with alcohol and placed in the box. then the box is aloud to sit alone for about 10min. uses lighter to lite tip of needle, heats till red, wipes down with alcohol cotton ball, then incs, then tapes holes. the jars are then placed into inc. chamber, 86d, that has been wiped down with alcohol and is sprayed with lysol all the time.

i get at least 20 out of 24 jars usually every time without even trying  :grin:

if the contams shows up at inc points, is the syringe contaminated, or could that mean you touched a contaminated object? i just done see why it wouldnt run down the side of the glass where the infected part would have touched as well. thats why i believe its comin from the syringe

I haven't lost a single jar out of my last 48 jars and i am a dirty son of a bitch. Guess I'm just lucky :laugh:
