Possible but challenging.
Much simpler to do Grain LC- I think there's a link to that somewhere.

the only way i see it not being a total fail is if you are talking about injecting some sterile water in a jar of fully colonized grain, shaking, and transferring the water to lc via a syringe.

You opened this jar, removed the grain, and want to use a leftover kernel to start LC?

Did you open the jar in sterile conditions?

It's a gamble, but a cheap one. Agar would be the right way to do it, but you might get away with LC.
Worth a try, I guess.

Edit-
Should have made Grain LC, then let it recolonize for spawning. Too late now.

Yes it can definitely be done, done it many times, there is a better way tho and a write up on how to do it. You really should have the colonized jar shake it so that the rye is all broken up and inject sterile water into the jar shake again and then suck up the water.
