I've never made a bottle, just jars, and whatever air gets in is sufficient. I haven't ever had to open my honey water up, except to remove the tag over the hole to get more in my syringe.
I do find, however, that when the mycos first start to grow in the honey water, they collect at the bottom of the jar (gravity). This usually occurs 2-3 days after inoc. I like to mix mine around a little, with only enough force to get the matter mixed in with the water more. This prevents 'clumping' that will start to occur about a week after inoc, which causes problems if you have a narrow gauge needle.

No air exchange is needed. Cover the hole with tape and then cover the lid with foil. If left undisterbed to incubate, the spores will develope small balls of mycellium around the bottom of the jar. Once you see these you can shake the jar and break the mycellium into smaller pieces. After about 5-7 days of incubation at 80-85F ou should have pretty good growth. Inject 10cc of H2o2 into the jar, retape the hole,(label)and cover tightly with foil. The jars can be stored in the refrigerator.
