isolation help - sectors, transferring w/ pics

This is my first try using agar/strain isolation. The petris were inoculated using a spore syringe, because I didn't have any prints to work with at the time. This is probably evident looking at the plates, because of the fact that its not growing out from a single point (my agar was slanted because I realized my glovebox isn't flat, which has since been fixed).
First things first - are these ready to be transferred to another dish? I'm very excited and can't wait to transfer them Its been 8 days since they have been inoculated.
When looking at the plate labeled "Mazatepec B1", my intuition would tell me transfer the mycelium from the 3 o'clock, 9 o'clock, and maybe 5 o'clock (???) positions. As you can see on the side of the dish, there is another white spot. I am almost certain that this is cubensis mycelium, since I used the spore syringe and the liquid tended to travel to the side of the plate. There is also some on Maz A1 as well.

Mazatepec A1 seems a little harder. It all looks pretty wispy to me. Any ideas of why it looks much thinner, with those circles in the middle? I'm thinking it might have to do with humidity inside the plate.
TC D1 is also pretty hard for me, as nothing really looks rhizomorphic as it does in Maz B1.

I *believe* I once read RR say that you might not see sectors until you do the first one or two transfers. Should I just take some of the more opaque "strips" and transfer it? Where would you guys transfer from? I have 4 petri's of each substrain, with three different substrains going. These seemed to be a good representative sample
I have both of RR's DVDs, but his plates don't look a lot like mine do. Thanks a lot!

Yeah if you can't see defined sections just take a bunch of samples that you think might turn out good. Once you see the sectors just take one or more samples from each and repeat until you no longer see sectioning.

from the 1st one i would take transfers @ 3 and 9 o'clock where the more stronger myc growth is at, and 12 and 6 o'clock where the more even growth is at. you have over 5 sectors in plate one. the second and third, i would wait a day to get more defined growth.

Yes, transfer the obvious sectors to new dishes now, and then transfer again as those sectors differentiate into more in the new dishes. RR

Five sectors? I am under the impression that there are too many sectors to see at this point. If there were only five, then my next one would easily be a single sector isolate.
RR: When you say obvious, I assume you mean the ones I pointed out. What about the plates that aren't so obvious (to me)? Thank you.

This is my mazatapec when isolated.I have lablled it M1 I would advise you to let it sit for a few more days to get defined sectors. I always wait until they get a little bigger. Good luck!
