petri 2 syringe

i have some fully colonized petris and i was woundering if any of u guys have an efficient method of transferring the mycelium to syringes, braking it up to as little pieces possible...
how to i get it out without agar getting into the syringe clogging it up?

cut a wedge and transfer it to either honey water/ or directly to your jars!
with the honey water, i'd let it sit for a few days, then shake the jar and suck water in your syringe!

You can't transfer solid pieces of mycelium to a syringe, as they're SOLID. I think the only way is via the honey tek.

Sure; A good way to do it is with a sterilized swab. Unfortunately most of the ones you buy in the store have plastic, and won't PC, but you may be just fine using the plastic ones after soaking in some alcohol.
Some things to watch for. . .some petri cultures are tough, rhizo, grow very flat and adhere tightly to the agar surface. Swabbing doesn't work as well with this type of growth. Most of the time there is some material that a swab will easily pick up. After swabbing up as much as possible, submerge in the solution you're going to make the syringe from, and holding against the side give a good twist. . .or otherwise dislodge the mycelial fragments. You can make a few repeats of this, and will generate tiny hyphal fragments that will go through an 18.5 guage needle just fine. If there are larger pieces present in the final solution, just let them settle a little bit and suck the top portion only. It's amazing how little visible material you actually need to make good inoculum.

Simple. In a glovebox or under a flowhood open the plate up, and inject an amounts of sterile water from the syring into the plate, scrape the surface of the plate with the syringe tip, then suck back up. trust me your syringe will be full of mycelial fragments.

This is correct and it works well. You have to be careful not to tear up the agar too much or you will plug up the needle. You also need to be 100% sure that there is nothing nasty growing on the edges of your plate or you will pick up the nasty as well, if the water makes it that far.

Ah yes, I should have noted that with the swab thing. . .often people formulate agar that is 'soft' so that even light swabbing will pick up chunks of agar. Agar will plug a needle quicker than anything.
I don't actually work with syringes I make screw-cap tubes that I use to directly inoculate grain jars; in that case it doesn't matter if there are huge chunks of anything in there [I make them *heavy*]. For working with syringes, the squirt and suck method may be better for you, as it involves fewer materials and steps, and will generate smaller fragments. Best of luck;

Just don't push all the water out of the syringe before you suck it back in. If you plug up the needle, just push the shit out with a bit of the extra water and then start sucking the water back up again. I never had a problem, but if you get a really bad plug simply heat the needle up in a flame melting the agar/plug and then squirt it out real quick.
It sometimes seems like your not getting anything done, but if you look close you will see small white flakes floating around. For BRF jars this method can take colonization times down to a quick week.
