Alternate to petri dishes!!

Hello ..

I found some agar, but canot find petri dishes,, i'm sure one can use something else... but what??

I was thinking 1/2 pint wide mouth jars if i can find them???? or maybe glad plastic containers.... Hmmm...

what else do you guys use???

When I didn't have petri dishes, I was using 125ml mason jars... It was easier like that. Prepare the agar, pour it into the jars, then pressure cook. :grin:

Question 2.. how think do you pour them? 1/4 inch or so sound ok?

pros and cons of thick or thin agar???

I would pour them thinish, there's no reason to pour them thick, if you pour them too thick, and end up using dishes later, the wedges will be too thick for the dish, and hit the roof.

When purchasing prepoured petri plates, there is such a thing as 'thick pour'. They pour to roughly twice the normal depth. This is used for organisms such as Mycobacteria and even fungi that could take months to grow. Actively growing cultures most often need air exchange [can't be parafilmed] and will tend to dry out after some time. The 'thick pours' last a lot longer.

Other than making the plate last longer [which can be accomplished sufficiently with a thin pour--parafilm, mineral oil, dH2O, etc] there is no reason to go very thick. Sometimes you might see mycelium race up the side of a rye/birdseed jar, above the level of grain. . .somtimes all the way to the top. Some of the nutrient sludge can form a film on the inner glass surface, effectively making a 'very thin pour'. You don't need much. Use the extra agar to make more culture jars; that is a more important thing when you first start out. . .a couple rep's of the same thing.

I found the perfect container yesterday.. they are made by " Ziplock ".

Round containers about 5 inches wide, 1 inch tall, they come with lids, are microwavable, dishwasher safe etc..... and cheap.. 1.65 $

Instead of having to whip out the PC,, i'm going to try microwave sterilizing in a glass jar, then transfering to containers.!!

ok i know this is a day late and dollar short, but i think its kewl.
sure, small 1 1/2" x 3/4", but they are threaded .1 screws into the bottom of the next . see what you find when you hang out at drug stores ? :grin:

Ignore me, somehow I got switched to threaded view and didn't see these other replies. Hmmm wonder how that happened?

Anyway, use baby food jars if you have them.

For depth, especially in 1/2 pints, most canning jars don't have a flat bottom. I pour so that the desired depth (1/4 in or so) is the depth in the middle of the jar, not the sides. Otherwise the agars too thin in the middle.
