Where do most of you do your agar work?

I want to start doing some projects with agar but I don't have the means to construct a flowhood. I was wondering if you all do your agar work in front of a flowhood or in a glovebox? I don't see the point in wasteing my time starting these projects in a glovebox if they are most likely going to fail. Sorry for the noob question. MM

With good technique, a glovebox will work fine. And a flowhood won't save you from poor technique.

You will have less succes with a GB but if you sterile procedure are good you should have pretty good result. I just don't like to do agar in a glove box but thats me.

I have had great success w/ a clear rubbermaid w/ 2 holes in it. I just set it on a little table in the living room and work in front of the boob tube.

I think my sterile technique is good. I very rarely get any contam. Guess this is a good way for me to test myself and my technique. MM

I too have a Question in which the OP will benefit from, Is agar more sensitive to contams then say PF TEK and or Grain work?
Reason I am asking this is like the OP said,I very very Rarley have a contam but have yet to work on agar for the fact that I was under the impression that agar was extremely sensitive to contams.

It's not extremely sensitive. I've had a stack of agar plates sitting in the back of my glovebox uncovered, with no cover over the arm holes for 36 hours before. Came back to the stack after running errands, living life, etc, inoculated and didn't get a single contam due to it sitting.
The reason it's more sensitive is that you have to open them and expose the entire usable surface area to the elements as opposed to injecting in a tiny hole creating a minuscule opportunity for contamination. Also they don't seal air tight like jars, but if you wrap in press n' seal or parafilm that shouldn't be an issue.

I don't see why mold spore would germinate more or less on agar. They will probably grow faster but who care. I may be wrong but im pretty sure its the same same thing as the other substrate. The difference is in the way to do it, There is a lot more chance that you intoduce contam in your agar while doing it cus of the way you do it.

My FOAF does his in any room without a draft. Meaning anywhere, just close the windows and make sure the air is not moving.
Barely gets anything contaminated.
He does wear gloves and a mask and sanitizes with alcohol and of course flame sterilizes with every move.
For the non-professional, he really doesnt think flow hoods or gloveboxes are necessary. When the time comes for you to build or buy one, you will know.

glove box at least in the last month i have done almost 100 dishes i have lost 4, 3 were bad cultures to start with and one was my fault

How about this...
get the agar and the dishes, work with them w/o a box or a hood.
see what you get.
if it contams, then get one or the other, but my bet is you wont need to.

I actually placed an order today and will have my dishes very soon. I plan on useing my glovebox. It seems that this whole project is going to be far cheaper than I had origionally anticipated anyway. Thank you all for the info as always. MM

I hate working in a glovebox, I find it very akward and uncomfortable. But then again i made my glovebox like a retard. I put the holes on the 2 sides so I am hugging it whenever i use it. Needless to say i try to use it as little as possible. I will be making a new one one of these days, and I would recommend putting the holes on the same side, like noobieshroomie's happy glovebox

yes, i feel like an idiot too Also, the arm holes are a bit small and its a pain to squeeze my hands through. Don't build you glove box stoned :P I put them like that so i could reach every point in the box, i think...
