The Cabinet that Should Not Be!!
    #11737482 - 12/29/09 01:27 PM (15 years, 22 days ago)

I need to make 50 posts to be a real boy

And so I thought I'd show the cabinet that should not be!

It's not the best setup.. but I'm improving it as I can.
Working on malt/dext LCs at present.. with a bag fruiting coming up once my bags arrive.

I'm a newb at LCs and figured I'd do brute force in the hopes of getting some that were not full of FAIL.

My daddy always said if you can't shoot straight you better have a machinegun.

Hopefully good tools will be joined by good skills in time :crazy2:

lemme be the first to say i like that ! +5

just do not let yourself get folled by "i have a hepa now i may work sloppily"
see it as another reminder how much sterility and aseptic working drills are important in this hobby

You probably can't see agar's LC post because of your post count so here's the full thing:

Quote:

agar said:

Pint jar on right is multispore inoculated (SA).
Allowed to colonize near 100%.
Syringe is 60 ML w/18 gauge needle.
Quart jar on left is sterilized water.

Aspirated 60 ML sterile water into syringe.

Injected 60 ML sterile water into colonized grain jar.

Shook grain jar to liberate mycelium from grain.

Injected total of 180 ML sterile water into grain jar.
(shaking mildly between each injection)

Aspirated out mycelium laden water from grain jar.
(all I could aspirate out of the grain jar was 150 ML)
Then, injected mycelium laden water back into sterile water jar.

Approximately 500 ML of Mycelium laden water.
(enough LC for approximately 50....10 ML syringes)

The advantage of this procedure is that you can be assured.
The culture in clean.
Because you can VISUALLY inspect colonization in the grain jar.
(if there is FUNK, you can see it )

That is not the case, with sugar type LC's.
(multispore syringe inoculated)
They can be contaminated, from day 1.
And, you NEVER know it. :frown:
Until you inoculate spawn jars with that LC.
Then, have every single spawn jar GO SOUTH. :mad2:

Sansa,

Thanks. That's very informative. I will try to get up to 50 so I can read all the content.

Bro,
It's the Series 1 portable from Fungi Perfecti.

"The Series I Hood
FP's least expensive Laminar Flow Hood now comes pre-assembled! All you need for final assembly is a length of electrical cord for the fan, a screw driver and caulking gun. This unit comes complete with a scanned, 99.99% efficient 24 x 12 inch HEPA filter, fan, 5-ply birch housing, screws and brass handles. Ideal for "on the road" tissue culture or for use in small clean-room environments. For those who have a limited budget, the Series I is uniquely affordable.
E-ALFH1"

I just took the handles off the side so it would fit in the used TV cabinet. That cabinet has a slideout tray for a big cathod ray TV.. which gives me a worksurface (suitably draped in plastic and bombed) in front of the hepa. It was a great find, got it used from the kinfolk. The whole thing can be locked with a wire bike lock so I don't have any problems with securing my gear.

It's not that hard. It would have saved you 300$ at least. Oh well at least you have one.
