hi, i've got two 10ml syringes left i'd like to create an isolate, and grow some more cakes too.
what's the best way to proceed?
my first / only grow was some pretty good cakes in a S.G fruiting chamber.. but i really want to do a bulk grow with an automatic fruiting chamber. but figure i need an isolate to do it justice..
thanks b

From MS to an isolate you have to do agar work

There are a couple ways. One: You can clone from fruits Two: Take spore prints and germinate on agar.
Cut out the best looking mycelium and transfer to new plates. Then inoculate grains.

How do you figure that one. If you have a fruit body that you like and has all the characteristics you want, cloning it isolates that set of genetics. How do you think commercial growers do it?

Mushroom tissue from ideal mushroom. sometime has other genetics still in it thus making it NOT a true isolate.
There is a difference from isolating and cloning

True, but it's a step in the right direction, and you can do a true isolate on agar from a clone.
"How do you figure that one. If you have a fruit body that you like and has all the characteristics you want, cloning it isolates that set of genetics. How do you think commercial growers do it? "
Cloning will still give you multiple genetics, but a smaller subset.

Chapter 14: Evaluating a Mushroom Strain <-- that's how commercial growers do it. A fruit body produced from MS inoculation will still contain multiple strains. Shea25 is right. You have to do agar work. Germinate the spores on agar, transfer sectors to plates until you have isolated a strain, then grow out and evaluate every strain. Take notes the whole way through the process.

Yea thats what im saying he asked how to make an isolate. cloning wont give you that. Its true that it will be better then MS but its not what he was asking for.
Clone= still multiple sets of genetics Isolate= Single genetics

I was meaning take the best looking mycelium from the clone and transfer as well as from spore germination. Forgot to separate that part from the rest. It all comes down to the mycelium that has favorable traits. I am not agruing your point shea, just trying to fix my typing skills.

Ah Ic ic . Yea you can do isolate from tissue. It would be easier to start with a sterile spore syringe if its someones first time. I personally like to use tissue myself

I have had great success with LC tissue cloneing. I am not sure if it is considered true isolation but it does help atleast. Well it has for me atleast. MM

Clone LC's are not true isolates. Also with LC's you never know if its going to be good or not. thats why agar is better

thanks
so i'll get some agar, and dishes..
and in a nutshell; inject ms solution {sterile syringe} wait for mycelium growth slice off a healthy strand make a master (isolate) grow isolate to test its merits..
^ right track?? would someone mind posting a link for this method
sorry if i seem a bit clueless.. but i did some thorough research on isolates/agar, and then promptly forgot most of it.
