Germinating Spores

lately, i've been trying to make my own syringes from prints, and then inoculate a batch of cakes, as opposed to buying more syringes, because i need to learn to be self sufficient, however, i havent been having much luck.
its not sterile procedure that i lack, i havent ever gotten any contams, during colonization.
anyways, this is what almost always happens. i make the syringe the usual way (sterilized shot glass, scrape spores in, shoot about 4-6 cc's of sterile water, swirl, suck back up) after this however, i never see any spore clumps in my syringes, but i mean i know they must be in there. so i inoculate a few jars, wait weeks, and weeks, and nothing happens. no contam... but no germination either.
can anybody tell me what i may be doing wrong when making my syringes?

If you're using a needle tip to swirl them you may be damaging the spores. Try using an inoculation loop-shaped piece of wire.

yes however i wasnt able to really suck them up well. after i suck up the spore water, there are usually still some clumps left in the shot glass

and you dont always have to see spores in the syringe for them to really be there. they are microscopic

did you make LC with those 3 different prints? did your spores come in direct contact with any chemicals -noobie-

Do you think it's possible that the shot glass still had rubbing alcohol pooled at the bottom when you scraped your spores into it? That's the most plausible explanation I have, so thought I'd ask. I always worry about this myself since I use the same method to make syringes. (No problems here though.)

well the other day someone had the same problem and it turned out they were pressure cooking their syringes with the spores in them.
it took 4 pages of responses to figure that out, so in the interest of getting to the heart of the matter; are you doing that?
