what's going on with these jars!!?!!

well after a 2 year hiatus from growing I decided to give it a shot again. I am now on my 2nd trial and still no luck. Ive since ordered fresh spores and ready to give this thing one last go.

Anyways, I cant for the life of me figure out where I am going wrong in my procedure. And both trials the jars havent contaminated, they simply dont start colonizing. I am using Tyvek envelope as my filter, with micropore tape covering the injection ports. I was figuring it had to either be a FAE issue or water content/ WBS issue.

For the WBS ive been soaking for 24 hours, then draining for about 30 minutes and loading it into the jars. been PCing 90 minutes at 15 psi

Anyone who can offer some hints, I appreciate it. Spores were purchased through Hawkseye. New batch through Sporeworks, so I will keep it posted.

From spore, it can take over a month to germinate, or less then 2 weeks.  Are you throwing your spawn jar contents away prematurely?  If its not contaminated then just let it sit!  Whats the harm in that?  How long has it been since inoculation of your WBS?  It'll help others to know your growth parameters as well.

even with no air exchange the spores will germinate and colonize well before stalling from lack of gas exchange.

So, either your wbs is way too wet/dry,

Or your innoculating too early after pc'ing and killing the spores with the heat

Or a heat issue with where your putting your jars to colonize (too hot, too cold)?

Or of course some sort of contamination....but i doubt this as you would at least see some active myc.

Need more info on your setup.

in my experience i find the jars are often too wet than dry..but that's just me

It's been 9-10 days since inncolution. I am not planning on tossing the jars. But I will monitor them in the next week or so and see what happens.

First run I let them sit a month and never saw any signs of growth.

I could have innoc'd too shortly after PCing, I do get a bit ansy and innoc when the jars are still warm. So I will make sure to let sit overnight or something next time

I've seen a few jars start after 10 days of nothing (incubated at ~80°F). A week later, they were almost 50% colonized. Nature can work in strange ways :shrug:

My guess was that the syringe was not shaken enough for that one jar's inoculations (best to shake syringe after every jar because the spores can re-clump in the syringe, especially if it's cold). So maybe the jar did not receive enough spores to colonize rapidly.

Im guessing thats your problem, but 10 days isnt all that long, certainly keep them incubating and start some fresh ones

yeah i have some grain that is really dry and im just now showing some growth at the bottom of a couple.
those were done on thanksgiving

You might have the answer there!!  But wait and see as it hasn't been that long yet. I did the same and innoced too soon and  those jars failed. Wait over night is best so PC in eve then leave them. Tin foil off though :smile:

Subject: PESA attempt Finch Seed

Soaked Finch Seed for 16 hours roughly with gypsum.
poured boiling water over seed.
Let it drain for a hour could maybe of left it another 1-2 hours, but the seed had no standing water on it.
filled jars too roughly 2/3rds full , one jar had 10ml of water in the other had 20ml next time i would like to try another with no extra water.
PC them for 1.5 hours could maybe turn the heat right down next time, had to put jars on sides as 1000ml jars were too big for PC.
Need bigger one!
Could only do one jar at a time.
Was again impatient and only left the first jar to cool for 5.5 hours and the second for just 4 hours if one doesn't germinate i will no why ! too hot!
Inoculated 10cc in each one down middle then roughly 3/4 round the edge but i struggled to get it there as needle was short. So might of failed at that. Also the the box i did it in is too small for 1000ml jars couldn't get syringe at right angle etc
Need bigger box!

Now in incubator at 81-84 depending on if the heating is on.

Fingers crossed !


After wondering why nothing has happened after over 2 weeks I reread this and saw i only waited 4-5 hours after PC!

Im still going to start that 2nd batch just to see if I can refine my technique a bit more.

also, simmering your grains will help get more moisture inside the grain, as opposed to just soaking.

I think I did jump the gun. But the 1st run with spores from Hawkseye got me nothing. And I actually let those jars go for a month. And it's looking like maybe 6-10 out of the 14 of these are going to colonize. But we'll see, I should have a much better idea before I take off for my trip on Thursday. Then they get to sit undisturbed for another 12 days

Just returned home from my trip. Like a dunce I left my window open and my place was pretty cold when I got home

Upon checking on the jars, the same about 6-8 of the jars have developed small clusters of mycelium. I went ahead and flipped the jars upside down and knocked the birdseed around a bit. Hopefully they arent going to stall because of the cold.

These jars were inoculated on Dec. 1st and I would guess the jars are only about 5% colonized

Hopefully this isnt fail

OK, when I go to spawn my grain to my substrate...

The holes on the monotub; Do they get filled with polyfill, covered with duct tape, or covered with tyvek, or D) none of the above?

You want to block off FAE.
I put in the polyfil when I spawn, so I don't have to mess with it later- then put the whole thing in a bag to colonize.
Should be enough O2 in there to last through the spawn run.

some things IME that caused it to not germinate

-not letting the needle cool down after flame sterilizing and before injecting
-grain too wet
-spores from spore bank (4 syringes that didnt germinate in a row)
-Innoculating jars before they have a chance to cool down from PC
-If you can't get your grain to drain dry, throw a teaspoon of vermiculate in the jars and shake. don't put too much in, it will make it impossible to shake later.


never had any luck with WBS, always turned to mush. try popcorn. (bags of jiffy pop - 1.50, one bag does 3 qarts) simmer for two hours until it's soft all the way through.

They ended up all fully colonizing, the cold just slowed them down a bit. Ended up taking about 6 weeks

My buddy just finished casing the two monotubs when I was over there earlier, hopefully he gets some decent results

The two tubs were cased with 2 different types of Coir

one which was more like bark chips and another that was more like a soil. so far only the soil one has a spot of myc popping through. The one with bark chips doesnt seem to be creating nearly as much humidity and may need a spray down

Im hoping that after about 10 days of colonizing they will be ready to fruit. Fingers crossed

Both tubs were misted today, the one that was a bit slower is still colonizing in the closet for a couple more days. The other one had a couple patches of myc poking through so It's now fruiting

Hopefully in 2 weeks time I have some pictures for you guys
