can you pressure cooker sterilize twice?

I pressure cooker sterilized some jars last night. I am wondering if I can add a little more water to the PC and sterilize for another 90 minutes today before inoculation without harm to the cakes. This only my 3rd or 4th grow and I have lost alot to contams, this time I want to take every precaution to be sterile. I baked my BRF and verm. for 2 hours at 275 degrees prior to mixing together with distilled water and putting in jars, and made my kitchen into the most sterile environment I possibly could without buying a HEPA filter. I would like to pressure cook em again just for peace of mind even if it is not necessary.
thanks for any input or tips on sterile inoculation I may not know.

No I didn't use a glovebox before, I tried to make one last night but I cracked the clear plastic box trying to cut the holes for the gloves. Do you think I should inoculate in the bathroom instead? I bet the kitchen air is worse. I planned on mopping with bleach and spraying the air with lysol and cutting off all air-flow to the kitchen before opening the PC.

Just get a plastic box, lay it on its side with the open top facing you, and tape some large oven bags over the top of the opening so they hang down. Reach in under them. Spray it down with lysol, wipe, spray again and let it sit for an hour or so undisturbed before working. Then wipe once more with lysol to remove any settled dust/contams and work inside it. Moving air is the killer for contams. Wear rubber gloves, rub them down with lysol or alcohol while inside the box and start your work. It sounds worse then it is, and you'll be amazed at just how these little things can drastically improve things. No need to cut holes and install gloves and all that. Even a cardboard box with aluminum foil taped to the inside will work.

As long as the jars are sealed or have filter discs on them, you can open the PC in the open air in your kitchen without much worry. Just wipe everything really well before and after putting them into your makeshift glove box. Flame sterilize any syringes before use, inside the box.

U can try using oost air disinfectant stuff, u could also try inoculating infront of the oven at a low temperature to creat a warm air updraft - this is what i have been doing and so far, i have only had 1 contam out of 7 jars, but this is still not an ideal sterile teknique.

A glove box would be the best option.

flame sterilised the needle and alcohole wiped it yeah?? between each jar??

I'm curious, too.  I'm doing the BRF PK Tek.

It was getting very late last night when I did made the cakes and put them into the pressure cooker.  I let sterilize for an hour and it was getting VERY late.  I couldn't sit around and let them cool.

Should I inoculate them or sterilize them a second time?

Is it damaging to sterilize them again?

no need to cook again
