Agar with no flowhood?

Can I pour agar plates and innoculate agar plates sucessfully using the oven tek? Do you think my contamn rate will be high? Is a flowhood required? The reason I asked in this forum is that not many people use agar in the cultivation forum.

Don't use the oven tek, it's worthless. You can successfully pour peroxidated agar in the open air, but it's better to do it in a small room with lots of bleach/alcohol/lysol/whatever sprayed around.

i agree with zeta, no (huge) problem with perox agar. since this requires mycelium, not spores, all (or most) contam issues are then pushed back a step, to spores germination. for this, i have found the cardbord spore germination method the best

Well the oven tek is worthless for making syringes but have had 0% contamn rate. Ive innoculated both quart jars of grain and many pf jars with no contams whatsoever. My house is not sterile by any means and it works fine for me

I've had good luck with the oven tek and agar, maybe it's just luck. But it's worked well for me. All my contams thus far have been on the edges, from not using parafilm.

I don't believe in the oven tek bro.
I just clean my kitchen up....sweep the floor, then mop it.....bleach the counters....wash my hands with anti-bacterial soap....spray the air with Lysol. Wait a little while...then pour my plates.
I use electrical tape to seal them.

You think the oven tek does more harm than help?
I also do a thorough cleaning, as i have with all of my endeavours. I have not had too many issues with contams. I think that just keeping everything sprayed down with lysol, and mopping/sweeping a few hours before your work really helps more than anything else, that's what i've always done.
Only contam issues i've really had are a couple of greens on wild prints, and some greens coming from my sides, as well as some bacterial problems with my grains early on. I will definantly try electrical tape now, thanks!

From personal experience and experimentation... the oven tek in my house was a complete disaster. Other people seem to have good luck with it... I did not. Every single plate that I innoculated in the oven got contaminated. The heat convections around my oven tended to pull up from the floor, into the oven, then back out over my work area and up from there.
If the oven works for you, stick with it. If not, move on to a simple glove box.
Take a cardboard box, cut the top flaps off. Tape seran wrap to the inside so that only plastic is visible. Turn the box on its side and hang more seran wrap over the opening. Spray down the room and inside of the box with lysol a bit before you go to work. You can heat the needles outside of the box and bring them inside to cool down (keeping the flame away from the seran wrap!). Using this method I get nearly zero contamination. It is so cheap you can toss the box after you are done.
Be sure you wipe your arms and hands down with a strong alcohol solution before starting to work. Also avoid long sleeves when working. You can get a box of latex gloves at the hardware store cheap. If you use gloves, be sure to wash them with alcohol too (after you put them on).

I have a glove box but its only suitable for syringes. It is complete with permanant gloves and is completely sealed with a light. But its made out if a 31 liter sterilite container (small). The GF cleans the oven often and is a clean freak. Have had 10 pf jars last over a month that were injected with dead spores using the oven tek. Lysol is god. I was wanting to get 5 or so dishes and try to get a good pure isolate. Then go to grain then poo. I did the pf tek 1 time and all but 1 cake were thrown away(damn GF). I then stopped for 9 months and went straight to grain and poo. I can get started with 5 dishes and all the supplys for 36$ from myco suply. Check the vendor forum for the kick ass deal. Thanks RK I will try the electrical tape.

I use a clear glovebox that everything that enters is wiped with alcohol - as is the interior, then i spray lightly with lysol, wait five minutes and then i put on gloves, wipe with bleach and poor my agar no problems - ive poored around 200 paltes and never had a direct contamiantion - only from wild prints that im expecting to get contamination from. A microbiologist student did not believe me so did a test by leaving five agar plates open in the glovebox for a week - only one showed any signs of microscopic spore germination - the other four were quite interestingly very clean - well above what he would have expected.

Damn now thats a sucess story. I was thinking of building a small plexiglass glovebox and using it strictly for syringes and agar. But then again a flow hood wouldent be that much more expensive.

Lolol yea dumb question. The whole point is that warm 200 degree air is being blowed out onto your surface area. Contamns cannot fall into any of you jars or agar and contamn it. If you can get your microwave to spit out 200 degree air and you can work in fromt of it then yea. Youd have to be fucking einstein

what about poring the agar put an empty dish on top , nuke it ,then after it cools you could wipe down the outside with alcohol ?

i think the agar would buble if you werent carefull and stick to the petri lid But im sure nuking for short bursts would work ok.
