I want to do some isolates )After i get a hepa filter( and thought I'd ask a question about isolating.

If you made smaller cuts on some rhizomorphic mycelium. Would you have a better chance of getting a strong isolate by targeting areas with more hyphae? Or an area that shows hyphae with straight thicker lines? I'm new to this but can someone explain? I can't read a book right now and it took me 49 minutes to write this.

lol.. I have been taking long thin slices of rhizo off plates.. but I am thinking of moving towards small sectors. Though, my longer strands have worked out.. I am newb

I'm not talking about what i took. I'm just saying you should try to take single strands like there are in the top section. it has already been isolated

I know this is probably a dumb question, and im pretty sure I have the answer, but im not up to this advancement yet. Im just on LC's. But in that dish I asume thats agar. where you cut a piece of tissue and grow it in the petri dish? Then what do you spawn that small dish to a bunch of grain jars?Or whatever sub your using?

the wedge/sample you take from that dish can go straight to grain, straight to lc, or straight to another dish for further sectoring.

I hope you don't mind using your dish as an example but I mean cuts at these certain areas.



A: is the longer straighter connections
B: the densely populated area

You want to pull from the healthiest looking mycelium in the first few transfers. If you are using spores scrapped from a print, it's likely you will not see rhizomorphic growth just yet. Rhizomorphic growth usually appears at around 3 or so transfers.

Honestly though, if you isolate a healthy looking strain from cubensis mycelium, chances are it's going to please you. It's hard to isolate out a strain that doesn't perform great.

Get ready to go through a lot of petri plates. When you start seeing sectors, take samples from each and start isolating out dozens of strains. Then take the strains to PF cakes and decide which one you want to put to a bulk from there. Here's some plates that might help you identify sectoring a bit more.

Those pictures look great jitsu! The type of mycelium growth I am looking for is "straighter", which (I hope) should be a little faster in colonization periods. The first picture is exactly what I am looking for!





A: Being the straighter pathways
B: Curve/intertwined mycelium

I would love to know if these areas are I am targeting are worth the interest.

Yes, any or all of those would be good choices for further work. Try to transfer as many as you can to individual dishes and start growing them out.
