How many sectors do you start with?

In working to isolate strains, how many multispore transfers do you make?

This was my first time working from a print on agar.  I now realised I used tons more spores than I actually needed, so my mycelium is pretty fluffy with layers of strains(i think).

I transferred from print to agar on 4 separate dishes, because I wasnt sure if thaey would all take. 

Now, I have 4 separate dishes, and I'm wondering if I should just start with one or teo transfers, because I'm thinking that I may not even see sectoring in the next transfer. 


I'm confused of how many  transfers I should cut out of these trays.

any suggestions?


btw: print was from The Greak Oakridge Giveaway.


Another question:  To those who use Parafilm; do you have problems with it tearing where the lid and base overlap on the petri?

Three or four transfer are good but it all depends on how far you want to go and how good you are at transferring small sectors.

I would take two from each tray and keep going until I saw some rhizomorphic sectors. I think that it's really up to you how many more dishes you want to make from each of those.

I have yet to use agar to isolate, but I think that you could use the water dilution method to keep from having too much material to "clean" at such an early stage. It helps to shorten the road to a pure isolation that you want to keep alive.

I guess I'll find out how good I am at transferring small sectors when I do these..:lmafo:

3 or 4 sounds like a good starting point for transfers.  I think I like the bottom right one best.  maybe take two transfers from the 9-10 o'clock, and another from the 2to3 o'clock areas.  I think the top left plate is kinda neat, it resembles a mandelbrot; i definitely should transfer for that reason alone.



I want to isolate a few strains from this oakridge print, then grow them out on cakes or mini bread pans.  keep the best performers, then store it to a slant.  :shrug:


I'm ordering a case of 500 petris, but I'd like them to last a while.. right now I have about 40 petris on hand.
