glove box procedure

I was looking at a glove box tek on this site, and the instructions said to cut a square in the box and cover it with tyvek. What's the point of this square? The only thing I can think of is that it's for sterilizing the needle so you don't have to keep taking it out of the box. If this is the case do you keep the needle inside the box, poke it through, and heat from the outside? Or, do you keep the needle outside and poke through to the inside to inoculate?

On a related note, when you open the lid to the glove box to put the jars in, after closing the lid, do you spray oust with the jars inside?

circles are fine..

sterilize needle and place in the glovebox as it cools..

after that there is no need to sterilize between jars, just wipe with alcohol swab

Well, regardless oh whether or not to flame between jars, is that the point of the tyvek square?
