colonization issues (pic included)
    #11350384 - 10/30/09 09:32 AM (8 years, 1 month ago)

I am colonizing 15 half pint jars (Brown rice flour)and three jars, in specific, seem to be giving me problems.

The jar on the left, regardless of the fact that i had 4 inoculation points, only seems to be colonizing from one.
The jar in the middle... it's pretty clear what the problem is there - 0% colonization.
And the jar on the right has one really small and seemingly retarded colony.

Does this JUST HAPPEN? Can it be that it is growing inside rather than on the sides? And is it ok that only one side is colonizing - will it eventually colonize the whole jar?

Temperature fluctuates between 76f and 82f and they are in a dark place. Some jars in there are almost at 80% colonization. It's been 2 weeks since inoculation.

How long has it been since you inoculated?  The growth looks normal/fine to me.

could be retarded spores lol could be contamination i would keep those away from the other jars just in case. but let them grow and see what happens they might only be slightly retarded lol.
was this from multispore or LC?
looks like your on the right track though. good job

its quite normal to get some innoc points not germinating, but no problem, the innoc points that do take hold will colonize the whole jar...just give them time.

The jar with no growth is a bit suspect, does the substrate look identical in colour and water content (look wet?) to the other normally colonizing jars?.

But, sounds like youve got plenty of good jars there

I have the same issues.

First batch: 9 jars, all colonized from one inoculation point
Second Batch:  6 jars, 4 showing growth, 2 from one point, 2 from 2 points, 2 aren't showing growth at all.
Third Batch:  4 jars, all showing growth, 1 from 2 points, 3 from one point.

Mind you, each batch was prepared in the exact same manner, no variation in recipe or technique at all.  I don't get it.

Not every seed you plant in the ground will germinate and grow. How is this any different with spores? Sometimes you will have better germination rates than others. No big deal. It also took a month for my first PF jars to colonize, and almost an entire month to see pins... Cakes take longer.

Wow, a month for pins?  That makes me feel better; one of my cakes has been sitting for over a week and hasn't done a thing.  Some of the jars in the first batch are just now reaching full colonization, over 3 months after inoculation.

It doesn't always take that long, but with all the cumulative errors on my first grow they all added up. Plus you factor in the seemingly randomness of genetics and you never know what to get. The more I work with this hobby, the more importance I see in collecting good genetics and saving the masters for extended period of time.

3  months after innoc ss32?...thats  a long long time....what temps are they  in?

I get a little annoyed at my jars  if they arnt over 80% after 2 weeks.

~70-72, that's the ONLY thing I can think of that would have slowed them down, but even then, a few degrees shouldn't make THAT much of a difference, should it?  The last batch is colonizing much faster than the first two, and like I said, nothing changed.  They're PF Classic.  I don't know if it's  known to be a slow strain, but it has been for me.  On the bright side, they're potent as hell.  I ate 1.4g off of the one cake that has flushed and had a great time.

If you want faster pf cakes, one way is to use my plastic technique.

Takes about a week to get fully colonized cakes.  I did some 2.5 quart cakes in spawn bags recently, took about 2 weeks to colonize though.

i had the same problem.
only one inoculation point was colonizing, then it stopped and it contaminated.
however, if the myc is still colonizing steady then it is only a matter of time that it colonizes the whole jar.
your substrate doesnt look to wet judging by the pictures which is good.
if the myc does stall i would put them under quarentine away from the rest cuz its bad news.  but their are many reasons why the other inoculation points didnt colonize like the syringe u had or not enough innoculant or bad spores etc.

Couldn't the middle jar have a bacterial infection?

Same thing happened to me.  Some jars exploded with mycelium while others just grew a spot and seized to continue.  I think thats just what happens.  You can probably lower the percentage of jars that do this by considering how many cc's you inoculate, where you inoculate, your method of inoculation, incubation temp, and really any other variable you can think of will effect the way your mycelium grows.

I just birthed a QT I inoculated on 09/05, several other pints jars are on their 2nd flush! But watch em, I had several jars from 1 lc that looked normal just progressed differently so I quarantined, they developed pink/maroon splotches of some odd contam. Can't be too careful.
