Spore Print To Agar.
    #11338548 - 10/28/09 01:28 PM (14 years, 5 months ago)

Well this is my first time doing print to agar.. I usually just due live mycelium to agar, but I got a print I want to transfer to agar..  Well I did this, my question is how long should it take to see some growth in the petri dishes using prints ?

from my experience from 2 to 15 days.

i have a dish with p. colombian spores that germinated in 2 days, the a+ albino germinated in 3 days.

the others took between 1 - 2 weeks tops.

Ah, forgot to say how i do:

first i flame sterelize inoculation loop, cool it down in agar then softly touch the spore print, then touch agar. If u can see the spores u already have too much. Then i drop a drop of sterile water on the inoc point.

PS: i'm still in love with the japanese girl

Thanks.. I received a very nice print in the mail with a strain I have never worked with so I wanted to start work on it asap, but transferred it to agar instead of syringe, just to make sure it was clean.

I usually scrape a little off with a scalpel directly to the agar. Then I cut the agar slightly, where the spores lay. I don't add water. IMO theres enough moisture in the agar. Should see growth within three to six days.

Good luck with it!

All I did was scrap a little bit of the spores into the center of the dish and thats it cause there was extra moisture in the dish. I think it's been 3 days now.

You should see growth soon.

I should have mentioned that I keep mine in the incubator at 27C/80F.
