OK to melt the lid on an agar plate?

ok so if i have an LC syringe and an agar plate i want to start with it

cant i just heat the needle and melt thru the lid

squirt my drop of LC out and then cover the hole with tape?

I have thought about pre drilling the lids with a hole, and using a dab of silicon.. then I also thought about after it grew out, injecting sterile water in there.. shake it all about, and draw it back out.. ?

I just poured agar, 2 days ago, in a 1/2 pint with a drilled lid... hasnt contamed yet

Why go out of your way to mix and pour agar plates then? Just make a grain LC if you're not trying to isolate specific growth, which I think would be pretty hard done through the lid.

You could inject through the lid and try and suck up a plug of agar with the growth you're looking for, and transfer like that but I don't imagine that working all that well.

You can do agar transfers in a glovebox, as long as you seal them up with parafilm/press-n'-seal you wont get too many if any contams.

yeah but you can use them only for the first them. When spore spore germinate you will need to transfer normaly anyway so normal petri dishes will be ok.

say the growth is isolated.. do a final transfer of the isolate into the rigged dish.. Dish LC of the best growth..? I don't see why not.. if it doesn't contam in the process. Just like noccing up a grain jar with a wedge to make grain LC.. except that step skipped, not as exact.. but depending on how well you transfer what you want.. it could be pretty darn close. I have a PE I am working on.. all the newest transfers are 360 degrees of the same strong growth.. I would feel confident in any area of the growth.. or all of it (perfect guinea pig).


I am not saying.. that this is true or will work.. but I have thought about it. These are my thoughts.. and I will do it one day.  :hehehe:

I'm not trying to be snide, but I have to ask what is the point of this? Why not just lift the lid and place a few drops quickly. It seems to me that this would be much faster and controlled than what you suggest.

Everything I can think of that you would do with the agar involves transferring it to an exposed petri dish or grain jar. If you don't want to open it now, what about later when you transfer away from it? Just a thought...

.. so lets change that norm.. and not open it.. so its not exposed.. ever. All transfers (at this point) take place in sealed vessels, and not exposed..? Sound good..? Sound gay..?  "You never know if your LC is contamed until you use it." correct? You don't feel odds are better if the original noc came from a clean petri, and never got exposed?
I will find out, one day.
