Mo Easy, Mo Sterile, Mo Betta Agar Petri Dishes

Materials Needed: -1/2 pint wide mouth canning jars -wide mouth platic canning jar lids -drill or hot screwdriver -synthetic filter disks(i THINK 90mm ) -agar agar flakes -light malt extract -foil -micropore tape
Mix: 1.5g - agar 1g - light malt extract 50ml - h2o *can be multiplied to whatever
Jar Preparation: ( ) -drill or hot screwdriver a hole in the center of a plastic wide mouth jar lid. (i use one 3/16th in the center. works well)
Agar Preparation: (use some thing youll be able to pour out of once mixed, and something heatable) -measure in agar flakes -measure in lme -measure in water -heat (i set my agar pourer on a metal jar ring, and place into pot/water, and bring to a boil, then simmer till consistency of mix is fluid, and very little chunk left) -foil over the top of pourer(probably not necessary)
Petri Preparation: -pour hot agar mix(dont burn your fingers) into 1/2 pint wide mouth canning jars -place filter disk over glass rim -screw on lid -add a layer of micropore tape (also probably not necessary, but it give you somethin to write on ) -add foil to the top, making sure it covers the seals of lid/glass -pressure cook(if im doing petris alone, ill do 1/2 hour at 15psi, with grain loads for 1.5 and theyre fine) -LET COOL OVERNIGHT IN PC, unless you want a BUNCH of condensation in your petris -glovebox inoculate with whatever you choose
Note. ive never inoculated though the filter disks via syringe, but if you do, cover with double layer micropore tape or something similar to filter. Note. i forgot...
and after leaving control jars out for weeks, and seeing no contam, im pretty convinced
and after watching all sorts of other mycelial projects run rampant on them, im convinced
and after my order of 500 disp. petris never showed,...im convinced,
that these will be my petri dishes for many future projects to come.
Hope this helps some ppl out!

perfect visibility it breaths a little with the filter
the only way theyre bad condensation is if you put it in a fridge tooooo cold.

the idea is to be able to look down from above the jar/dish(better viewing angle), I have tried jars, and a clear lid is infinitely better IMO. But whatever works and definitely cheaper.

you dont need to look straight down.
you have high walls
one thing ive started doing to simplify and quicken transfering, is marking on the glass with a sharpie the "area" where ill be taking mycelium
that way i know right where its at and can open, cut, close
no tape
no parafilm
do it

yeah I use the marking trick too.
another neat trick: shine a flashlight up from underneath the jar, it might help you locate desirable areas more easily... (the ropier strands will block more light)

yeah, ive got some sick lil work lights
i think one of them i found was made SPECIFICALLY looking thru petris
but fo serious all, im thouroghly satisfied with these petris
always heard about the non-disposable petris being a pain to clean, and they get gnarly after a while, well these are SO EASY to clean, as anyone whos used a 1/2pint jar for cakes would know.
and the fact that youre sterilizing AFTER pouring the agar, nearly 100% guarantees its sterility until you inoculate, or open.

Sweet. I was just thinking today that I was going to order up some agar tomorrow. I've seen people do this before, but forgot. That is, until I saw this thread. Thanks dude!
Peace.

So are you back to 1/2 pints or recleaning those couple sleeves?
And dammit, I have to wait another day to dive in! I tracked down LME, but none of the grocery stores around have agar... I'll have to widen the search radius tomorrow... Just another version of the patience lesson to chafe my nutsack

patience grasshoppa! yessssssssssssss seems like after a year of not seeing pins and fruits, this tub is taking eons to fruit... hey i wouldnt try reusing disposables 1/2 pint jars, or real glass petri dishes can reuse for awhile worth the investment...? idk when you get going with agar, you all of the sudden are swimming in petri dishes. i imagine if i had enough money to buy enough reusable petris to support my habbit id probably pick up a worse habbit

Nice. Feet are getting wet.
Doubled your recipe and filled ten jars. That works out to approx 10ccs per jar. Seem about right?
Thanks again for the push. I'm looking forward to some mad scientist shite
