Transfer away from the contaims
    #11323301 - 10/26/09 10:00 AM (14 years, 5 months ago)

K so I got three plates that I would like you to look at and see if what I see is what you see.  Sorry the pics are crap.  They are two section plates a will be the top b will b the bottom then an hourly reference to see the sectors. for example a 1-2 would mean the top section sector is from one oclock(by my clock) to 2 oclock

APE

a1-3 a3-5 a5-8 a8-10 a10-1
b1130-230 b230-5 b5-8 b8-1130
Now for this plate I would think that a1-3 and b1130-230 are the most promising sectors

Next up an EQ plate

a1230-130 a130-3 a3-4 a4-530 a530-630NOTE a630-10 a10-1230
b12-3 b3-530 b530-630 b630-8 b8-9 b9-1030 b1030-12
I would think a3-4 is most promising and b8-9 b9-1030 b630-8 all look good to me with no real difference that I can see
NOTE I cut a transfer and moved to this dish

EQ second transfer

a  kinda looking like a monoculture
b1030-2 b2-5 b5-730 b730-1030

So what do you think are the best sectors and am I wrong in labeling them?

I can't make out anything from those pictures.  I'd suggest taking every growth from those dishes and transferring to new dishes.  You should be able to inoculate two or three sleeves of dishes.  Then, repeat the process in a few days.  Fruit every isolate to determine the best performer(s) and then go back to your well-marked dishes in the refrigerator to grab younger versions of the cultures that were best.  You can't eyeball the best cultures to transfer, especially on the first few transfers from spores. That's why I grab them all.

How the hell do you "well mark a plate" ive been thinking of this but dont have any solid ideas about doing this.  Thanks for taking a look rr yea i need a camera.  I knew that it is best to transfer all sectors and then do as you suggest but dam I dont have that many dishes and not too much agar atm

start with each transferable piece. label 1st one a1, second one b1, 3rd one c1, and 4th one d1

then when you transfer  from a1, label it  a1-2
so you'd know it was from a1-transfer2

put a slight scratch (if the petri is plastic) on the top and bottom to line the lid and dish back up.. circle the areas with a marker on the lid, and number.. or mark 12 o'clock with a marker on the dish, and use your 1,2,3,4,.. o'clock method.. or mind meld with the rhizo.. or become one with the dish.. or ask it

yea the mind meld why didnt i just think of that.  uhm my problem with marking the top is that when i move the plate to do transfers im going to fuck up and not match it back up again and i dont tape my plates closed i could but i dont.  The scratch is a good idea maybe on the side of the plate

yeah.. scratch the lid and plate..  would be easy to line that back up.. it would only become hard to mark the lid if you did a bunch of transfers without catching up first.. but I don't know, I haven't got that technical yet.. right now, if I see no contams.. then..

:highfive:
