Best way to transfer spores from faint print to agar?
    #11320118 - 10/25/09 08:07 PM (8 years, 7 months ago)

Hi, I have a faint lib print on printer paper.

i had three ideas:

1. just scrape the print and hope (assume?) many spores will land on the agar even though I can't see anything

2. cut a small piece of the paper and put that on the agar

3. wash a drop of water onto and off the paper, then put it on the agar

what should I do?

Can you get a swab or inculation loop?

damn that was a fast reply

maybe i've mistaken something for a long time. I thought you scraped the spores so they would fall on the agar, but using something to scrape the spores and then touch the agar - that must be the way to do it.

thanks

What I do is use an exacto knife and flame sterilize it, then once I think it has cooled off enough, I just scape a little and I mean tiny bit of the print into an agar petri dish.  It really doesn't take alot.

Also if you get contamination's, you just transfer healthy mycelium to a clean petri dish.

You can make a home made loop out of a pen and a piece of wire..  Just cut the wire about 2 inches long then stick both ends in the eraser end of the pencil, and BOOM a quick and cheap loop.

You can also cool your flame-sterilized tools in the receiving agar dish itself before making transfers or scraping spores.

Dip your inoculating loop into sterile water.  Place one drop of sterile water on the print, rub it around a very small area, and then transfer to agar.

I don't have a lot of experience with agar yet but definitely can just scrape your print then slice the center of the dish. I avoid water after many failed dishes.  I just can't afford a HEPA filter haha
