Cloning some Shiitake
    #11316656 - 10/25/09 05:51 AM (8 years, 29 days ago)

Just harvested my Shiitakes and now I want to do some cloning.

Thing is. I have some malt extract but it is hard as a rock. Would it still dissolve and be usable for agar?

Also. I don't have any alcohol or peroxide to sterilize the mushrooms. Is there any other way to ensure a sufficient degree of sterility?

yeah man cut into the inside of the mushroom and just use inner tissue.. as far as the malt agar.. it will work..take a drill and drill out what u need. mine is like this i have had it for years.. and it was given to me ,no telling how long they had it. i did a clone of a wild oyster about 3 weeks ago on some malt agar using some inside tissue. the first plate i took had no contaminates

Not sure about the agar, but I'm sure if you mix some hot water in to it it will dissolve.

for the cloning, you do not need to surface sterilize, just use tissue from within the mushroom, tear it open carefully and with sterile forceps tear a piece of tissue from the inside and place that on agar.

A substitute for peroxide or alcohol is a 2% bleach solution and soak for 2 mins, then rinse in sterile water.
