Extend The Life Of Your Honey/Karo LC Syringes.
    #11308949 - 10/23/09 11:53 PM (8 years, 1 month ago)

This is a work in progress but it just struck me.
I bet this TEK could make LC syringes last for twice or perhaps four times longer than regular LC syringes.

I'll add pics in the future. Comments and suggestions are welcome.

Step #1. Make your Honey/Karo LC as normal, let's say 100ML water & 4 ML Honey/Karo Syrup.

Step #2. Wait until mycelium cloud has consumed nearly all nutrients and liquid is almost, or clear. With Karo this is more difficult to distinguish.

Step #3. Make an equal batch of your Honey/Karo LC once more.

Step #4. Mix new LC into existing LC. Shake vigorously and begin filling syringes.

Step #5. Inoculate a jar of grains just to be 100% that it's mycelium that grew and not a contaminant.

Now the mycelium syringes go into the fridge to slow cell division, but the difference now is that there are nutrients available to keep the mycelium alive for a much longer period than if you made syringes with a completely consumed LC.

Example would make twenty 10Ml syringes that would last for 2-4 years instead of the expected maximum of one year.

What do you guys think? Is there a TEK like this already? If not then I'll make pics soon.

Warmest Regards All

Bit early to call it a tek, I think. Since right now it's mostly just some stoned rambling.

HeH!

No, it just struck me. I was thinking. If I clone a fruit some day in an LC. How can I make sure its shelf-life is extended? Then it struck me, give more nutrients right as you begin to slow the cell division process.

Yet current metabolism research points to calorie restriction as an avenue of research in the area of longevity.

And it's not going to grow in the fridge anyway- you're just providing food for contaminants.

Ouch, that's harsh, but yeah. Grain LCs are more stable because their are no nutrients. The myc enter a kind of stasis, or suspended animation. I've also heard of myc in oxygen free distilled water being kept for years at room temperature.

Well there wouldn't be any contaminants if you worked in a sterile environment.

You're right about calorie restriction, but complete restriction is death. Most people put LC in the fridge once the nutrients are consumed and I'm betting that's why it only lasts a year maximum.

Okay you made that post four years ago.

When you store the mini-LC's does the master LC still have nutrients in the water or do you wait until the master's cloud stops growing and then inject into mini-bottles?

Do you still have a four-year old specimen that you can try to knock-up some grains with?

Thanks for the input.

I had this same idea recently..

I dont see why you cant suck up steril honey water to refill your syringes.

I was thinking too, instead of making them into syringes just add an equal amount of fresh LC then put the whole jar in the fridge.

That would save a LOT of work. =)

Most people put LC in the fridge when they are too antsy to look at it any longer.
Taste an LC. Sugary, right?
That means there's sugar left.
Which means it's not all gone.
Which means you don't need to add more.

You are trying to solve a problem that doesn't exist.

Aha... here we go. Distilled water storage of mycelium has been an accepted method in mycology since the 1930s.

Resurrecting a Better Method for Long-Term Storage of Mushroom Cultures

exerpt:
"Screw the lid on tight and wrap Parafilm around the top of the vial to make sure it is airtight.

When you come back in a few years, you would not want to find that the water had evaporated.

Store the vials at room temperature away from direct sunlight. A bookshelf or wall cabinet is an excellent place. If conditions deteriorate, and the room should become unbearably hot, the vials can be refrigerated, but that is not normally necessary.

In the distilled water envirnment, the mushroom culture enters a dormant state, and it is held in stasis."

Hmm

They why do people here complain that LC's are only good for a year though? Maybe two?

And Doc.

The problem does exist, people here say it does...

Not trying to argue, but they claim LC syringes are good for like a year.

It takes a little more doing to make a pure DH2O culture than a karo LC. GLC will get you pretty close, though.

Doc said that running out of nutrients is not a problem in storing LCs, and he's correct.

I already see two things missing in the article...

Parafilm is breathable. And, have they tried to reanimate a thirty-year old vial? They only mentioned three years.

Because aren't spores similar to seeds.

Where a seed has nutrients inside of it to remain "dormant" without dying, but if those nutrients are used up, then the seed itself will never germinate because it's dead.

Aren't spores similar?

I didn't get the parafilm bit either. I've seen other examples with sealed vials, but I'm through searching this evening. Anyway three years, at Room Temperature? I think it proves the point. Try that with a Karo LC.

I remember RR stating that spores have an extremely low germination right from the start.

Is there any metabolic activity in a spore like a seed? What happens inside of a spore while it waits to begin germination?

If nothing is happening in there, then how does the spore "realize" when it is time to begin germination?

I don't know, but I doubt they're similar to seeds.  Fungi are a weird and remarkable life form.  Perhaps they are capable of starting metabolization of nutrients in the environment more directly.
